体质量调节对哺乳动物的生存适应具有重要作用(Kaplan,2000)。在许多哺乳动物物种中,体质量都有相对精准的调节,这种调节主要反映的是机体内能量摄入和能量消耗之间的平衡控制(Mercer,1998)。长期以来,动物的能量平衡和体质量调节机理被作为生理生态学研究的核心问题之一(李兴升,王德华,2003)。
近年来,由于生活水平的不断提高,肥胖问题在全球范围内已经变得十分流行(Farias et al., 2011)。而随着肥胖问题的突出,体质量、体脂的调控机理也逐步得到了深入研究(Hall & Guo,2017)。研究结果显示,肥胖主要是能量摄入、消耗和储存之间长期不平衡导致的结果,正常情况下,动物个体的体质量受到严格调控而保持相对稳定的状态,但是当调控系统被破坏时,能量的摄入和支出失去平衡,最终导致动物体质量出现明显变化(Tremblay et al., 2004)。
在对哺乳动物体质量调控研究过程中,相应的体质量调节模型不断被提出和修正,目前,主要的理论模型有4种,分别是调定点模型(set point regulation model)、适应点模型(settling point regulation model)、普通摄入模型(general model of intake regulation)和双干预点模型(dual intervention point model)(Speakman et al., 2011)。
1 主要研究进展 1.1 调定点模型体质量“调定点模型”是Kennedy最先提出的体质量调节模型,该模型的主要内容是:哺乳动物身体有一个内部控制机制,脂肪组织能够产生一种传递脂肪状态的信号到下丘脑,从而允许动物相应地调整其摄入量或支出,以保持脂肪储存的恒定(Kennedy,1953; Farias et al., 2011)。
20世纪90年代中期,美国洛克菲勒大学Friedman的研究小组发现了瘦素,瘦素是由白色脂肪组织分泌的16 kDa的蛋白质,由167个氨基酸组成,主要通过负反馈作用对体脂含量进行调节(Zhang et al., 1994)。这一发现将体质量调节机理的研究推向了一个新的阶段,为“调定点模型”做了进一步的补充,提供了强有力的分子证据,从而使人们重新认识脂质调节模型的重要意义。Tam等(2009)对这一模型以及瘦素在模型中的作用进行了数学上的推演和论证。
长期研究显示,许多哺乳动物在摄食、体质量和肥胖方面表现出明显的季节变化,但也存在一些小型哺乳动物在面对季节变化时,体质量能够保持相对稳定,符合“调定点模型”(Mercer,1998)。栖息于我国北方地区农田和草原的啮齿类物种黑线仓鼠Cricetulus barabensis的体质量并未表现出季节性变化(赵志军等,2014);赤腹松鼠Callosciurus erythraeus在季节更替的过程中也能够维持相对稳定的体质量(贾义平等,2017)。云南高原的昭通绒鼠Eothenomys olitor体质量在季节改变时变化不显著,符合“调定点模型”(杨押佐等,2021),而这一结果与横断山区的其他物种出现了种属差异,中缅树鼩Tupaia belangeri冬季的体质量高于夏季,可能受其分布区为热带亚热带和高原地区的影响,冬季食物相对充足(Zhu et al., 2012a);由于大绒鼠Eothenomys miletus、高山姬鼠Apodemus chevrieri和中华姬鼠Apodemus draco在冬季受低温胁迫较强,体质量显著低于夏季(朱万龙等,2008;Zhu et al., 2012b, 2013a)。小林姬鼠Apodemus sylvaticus以及位于以色列炎热的岩石沙漠中的金黄刺毛鼠Acomys russatus和普通刺毛鼠Acomys cahirinus等的体质量在全年无显著变化(Klaus et al., 1988;Kronfeld-Schor et al., 2000),也符合该模型。
除了小型哺乳动物以外,人类成年个体的体质量也能够长期保持在一个相对稳定的水平。比如在减肥结束后的一段时间内体质量出现反弹(主要原因是体质量减轻的人代谢低),这与“调定点模型”一致(Weinsier et al., 2000)。体质量反弹除受代谢影响外,还与机体能量摄入增加和消耗减少密切相关(Kasher-Meron et al., 2019),这也给当今肥胖流行问题和正在减重的人提供了参考,想要保持体质量在相对稳定的水平,必须严格控制能量的摄入和支出。此外,许多物种由于进食过多或者过少会出现增重或减重的现象,但是当刺激体质量变化的因素被消除后,这些动物可以通过下丘脑及时调节食物摄入或能量消耗,使体质量再次恢复到原来的状态(Anderson et al., 2001;Müller et al., 2010)。
从20世纪50年代以来,为了探索“调定点模型”,人们进行了许多脂肪切除术实验,但得到的结果却相互矛盾,一些研究显示,脂肪切除后的一段时间动物的脂肪能恢复到基线水平,另一些则没有(Michel & Cabanac,1999;Seretis et al., 2015;Murillo et al., 2019)。在对各种哺乳动物脂肪切除的研究中,如黑线毛足鼠Phodopus sungorus和草原田鼠Microtus ochrogaster在实验结束后体质量并未发生显著变化(Demas et al., 2003)。一项对大鼠Rattus norvegicus体质量调定点测量的研究表明,限制食物量在短时间内会降低大鼠的体质量,但不会降低其体质量调定点(莫文清等,2017)。
Levin(2006)提出,调定点可能被环境因素改变,特别是在发育早期。例如,尼古丁会降低雄性大鼠的体质量调定点,当体质量降到调定点以下时,这些动物开始囤积食物,当体质量恢复时,囤食现象消失(Frankham & Cabanac,2003)。对人类的研究也获得了类似的结果,主要表现为人的饱腹感会因为尼古丁的摄入而加速(Cabanac & Frankham,2002)。动物体内的某些激素也会影响体质量调定点,最典型的是大鼠在切除肾上腺后体质量调定点降低,表明糖皮质激素浓度和下丘脑促肾上腺皮质激素释放激素参与了体质量调定点的调节(Gosselin & Cabanac,1997)。
1.2 适应点模型尽管“调定点模型”为体质量调节提供了较好的理论基础,然而,由于其自身的缺陷,有学者持有批判观点(Wirtshafter & Davis,1977;Levitsky,2005)。主要是由于“调定点模型”不能很好地解释现今流行性肥胖的问题,而将肥胖主要归因于内在的生理因素(Tam et al., 2009),因此在此基础上提出了“适应点模型”:体质量只是根据生活方式对能量平衡的影响而稳定到一个新的平衡,并将动物身体能量存储调节类比为“湖泊的水位”,即在任何一个有输入(食物能量)和输出(能量消耗)的储存体(如脂肪储存体)的系统中,如果任何一个输入被下调,系统的储存体就会达到自然平衡(Speakman et al., 2011;Speakman, 2018)。
瘦素抵抗就是该模型比较重要的一个例子,Tups(2009)指出,瘦素抵抗主要表现出2种主要状态:一种是病理性,例如食物诱导性肥胖;一种是适应性反应,即动物为了应对环境变化带来的极端挑战,允许体质量的变化高于或低于调定点,比较典型的是哺乳动物的妊娠期或动物体质量的季节性变化。例如,妊娠期的布氏田鼠Lasiopodomys brandtii和大绒鼠为了适应能量需求的增加,其体质量调定点升高,可能也是存在瘦素抵抗(Zhang & Wang,2008;Zhu & Wang,2014)。此外,季节性哺乳动物也常作为研究瘦素抵抗的有力模型,当这些动物在面对光周期的季节变化时机体能量出现不平衡,需要及时调整体质量,最终达到新的能量稳态(Tups et al., 2004)。研究显示,黄腹土拨鼠Marmota flaviventris的血清瘦素水平在秋季体质量增加阶段也出现增加的趋势(Florant et al., 2004);黑田鼠Microtus agrestis从短光照到长光照,瘦素浓度升高,体质量增加(Krol et al., 2006);对黑线毛足鼠来说,光周期的季节性变化是影响其瘦素抵抗作用的关键因素,并且这个过程可能是由下丘脑GRB2蛋白减少介导的(Tups et al., 2012)。
调定点模型和适应点模型应用比较广泛,此外,还有普通摄入模型和双干预点模型。
1.3 普通摄入模型食物摄入量被认为是调节体质量过程中的关键控制因素,尽管动物在生存过程中的食物摄入量变化很大,但通过调整摄入和支出,最终也能够使体质量保持相对稳定(Chow & Hall,2014)。“食物诱导产热假说”也指出,动物在摄入过多的能量后体质量却不增加,主要原因是非颤抖性产热将机体过多的能量以热的形式释放了出去(Rothwell & Stock,1997)。
在哺乳动物体质量调节过程中,有关食物摄入调节的模型主要是“普通摄入模型”。该模型是在“调定点模型”和“适应点模型”的基础上提出的,主要内容是食物摄入受到生理、环境、社会、心理和饮食等因素的影响,而这些影响因素可以分为补偿因子(主要是生理)和非补偿因子(主要是环境)两大类,补偿因子与摄入量有负反馈循环,同时影响摄入量并受到摄入量的影响,而非补偿因子影响摄入量,但不受摄入量的影响(de Castro & Plunkett,2002;Speakman et al., 2011)。该模型提供了一个比较全面的解释,将生理和环境两类因素对饮食的影响整合到摄入和体质量调节的过程中,有助于将体质量调节与其他食欲调节相结合。
补偿因子方面,比如胃内容物和饥饿感影响摄入量,反过来又受到摄入量的影响(de Castro,2010)。食物的摄入主要依赖于中枢神经系统产生的各种刺激和抑制肽,促生长激素神经肽家族中的新成员alarin具有刺激动物进食的作用(Gregory,2012)。下丘脑中的神经肽Y和刺鼠相关蛋白具有促食作用;而阿片促黑色素原和可卡因-安他非明转录调节肽则具有抑食性(Tang et al., 2010)。下丘脑外侧的特定神经元产生食欲肽(食欲素-a和食欲素-b)控制食物摄入(Boss & Roch,2015)。胃饥饿素是一种在胃中产生的酰基化肽,通过与生长激素促分泌受体的相互作用,能够刺激动物进食(Shiiya et al., 2011)。下丘脑神经肽中的神经肽W也参与哺乳动物的摄食和能量平衡的调节,其与下丘脑内其他含有食欲调节肽的神经元一起构成食欲调节神经元回路的一部分(Takenoya et al., 2010)。
虽然这2类因子对哺乳动物摄食量具有不同的影响,但是它们是同时发挥作用的(de Castro,2010)。温度、光周期、植物次生化合物等非补偿因子对动物食物摄入量都有影响。温度对食物摄入量影响较大,像高山姬鼠和哺乳期的大绒鼠等在低温驯化后会增加食物摄入量,弥补热量散失,防止在冷环境中体质量过度降低(Zhu et al., 2011;朱万龙等,2017)。光周期作为重要的生态因子,对动物食物摄入的影响具有种属差异。主要表现为短光照会增加布氏田鼠和根田鼠Microtus oeconomus的食物摄入量(Song & Wang,2006;Wang et al., 2006);而中华姬鼠和F344/NHsd大鼠等则在长光周期下表现出食物摄入量增加(Zhu et al., 2013b;Ross et al., 2015)。植物次生化合物会影响植食性小型哺乳动物的食物摄入量,根田鼠和东方田鼠Microtus fortis等在食用单宁酸后的食物摄入量受到显著的抑制(李俊年等, 2003, 2007)。在对人类进食的研究过程中,发现进食环境可能会分散个体对食物的注意力,加剧暴饮暴食,扰乱个体对摄入量的监控;而社会交往也可能会分散人的注意力,从而影响摄食行为,人们在集体进食时比独自进食时会多摄入50%的食物(Ogden et al., 2015)。
1.4 双干预点模型体质量的平衡调节并不总是将调节变量维持在一个理想值,而是将其维持在可调节的范围内(Geary,2020)。因此,学者基于小型哺乳动物和鸟类的数据进一步提出了“双干预点模型”(Levitsky,2002;Speakman,2007)。该模型指出,动物的体质量不是由调定点控制的,而是由上、下干预点进行调节,将体质量调整到可接受的范围内,上干预点是由与捕食风险相关的选择压力设定的较低的干预点, 下干预点是由与饥饿风险相关的进化选择压力设定的,可以防止动物的储备量过低而陷入饥饿的危险,保证正常的生理需求,解释了为什么我们通常抗拒减肥(Speakman,2007)。因此,“双干预点模型”明确了体质量调节和环境压力有关:当环境压力增大时,动物会降低体质量(Speakman et al., 2011)。
上干预点是许多野生动物(包括哺乳动物和鸟类)体质量调节的关键(Gosler et al., 1995)。一般来说,更大的体型与更大的能量需求相关,为了满足生存条件,动物会增加觅食活动,导致被捕食的风险增加(Speakman et al., 2003)。被捕食风险影响着物种的生存策略(Navarro-Castilla & Barja,2014)。相关研究也显示,拥有高水平的脂肪会导致动物被捕食的风险增加,因为体质量较大的猎物可以缩短捕食者搜寻的时间(魏万红等,2004)。所以,捕食风险增加对体质量/脂肪有抑制作用,对小型哺乳动物的影响比较显著,如黑田鼠、东欧田鼠Microtus rossiaemeridionalis和小林姬鼠(Sundell & Norrdahl,2002;Monarca et al., 2015)。研究显示,野外生存的高山姬鼠可以通过调节能量代谢和产热来防止体质量的过度增长,达到降低被捕食风险的目的(高文荣等,2013)。
上干预点并非是一个固定值,Speakman(2008)认为200万年前,火的出现和社会的发展,使人类经历的被捕食风险降低,决定体质量上干预点的基因一直受到随机突变和漂移的影响,这种对捕食压力的释放使上干预点的位置向上漂移,而下干预点的位置没有移动。虽然觅食者因饥饿造成的死亡率较低,但哺乳动物仍会根据饥饿风险调节体质量(McNamara & Houston,1990)。像人类以及一些小型哺乳动物,如长爪沙鼠、金黄刺毛鼠和黑线仓鼠等在限食后的体质量和体脂含量显著降低(Xu et al., 2011;Ben-Zaken et al., 2013;Wen et al., 2018)。C57BL/6J小鼠在短期间歇性禁食实验中表现出,前期体质量有降低趋势,但后期又表现出体质量和脂肪增加的趋势,最后恢复到基线水平,主要是为了抵抗食物短缺带来的胁迫(Zhang et al., 2012)。以上结果与“双干预点模型”一致。但研究显示,即使当食物短缺的风险为0时,动物也不会选择将脂肪储备的水平调高,而是调整在较合适的水平,以维持生存(Higginson et al., 2016)。
该模型虽然具有一定的进步性,但只提供了动物体质量调节的2个相对的选择压力阈值,目前对如何精确定位上、下干预点还没有形成统一的意见(Higginson et al., 2016)。
2 结语与展望哺乳动物的体质量调控一直是动物学家们研究的重点。虽然经过数10年的探索和发展,目前已经提出了几种调控模型,但每种模型都有自身的不足之处,因此,需要在后续的研究中不断进行补充和修正,后续研究可以利用新方法、新技术从以下几个方面深入开展:(1)对于符合“调定点模型”的动物,可以研究在引入天敌、设置不同光照、食用高脂高糖食物后,体质量是否会发生变化;(2)确定和精确定位体质量调节的上、下干预点。本文通过综合研究与分析,为更好理解哺乳动物的体质量调控机制提供更详细和准确的资料,同时也为解决人类的肥胖问题提供参考依据。
