摘要:研究表明, 微塑料对生物体具有不利影响。本研究以剑尾鱼Xiphophorus helleri为实验对象, 将其暴露于不同粒径(0、1 μm、5 μm、1 μm与5 μm按颗粒数1∶1混合粒径)、不同浓度(0、1×106粒/L、1×107粒/L)聚苯乙烯微塑料水环境72 h后, 测定其消化酶、抗氧化酶及免疫酶活性。与对照组相比, 各暴露组肠道淀粉酶、脂肪酶和胰蛋白酶活性均显著升高(P < 0.05), 超氧化物歧化酶和谷胱甘肽过氧化物酶活性显著下降(P < 0.05), 过氧化氢酶活性显著升高(P < 0.05), 丙二醛和溶菌酶活性显著升高(P < 0.05)。微塑料急性暴露会干扰剑尾鱼对食物的消化吸收, 并影响其肠道抗氧化酶和免疫酶的活性。

Effects of Microplastics with Different Particle Sizes on Intestinal Digestion, Antioxidation and Immune Enzyme Activity of Swordtail Fish

Abstract: Plastics are widely used in many fields because of their stable physical and chemical properties. However, the annually increased global application and output of plastics have already led to the continuous accumulation of microplastics in the whole ecosystem. A large number of studies have shown that microplastics have adverse effects on organisms. In this study, swordtail fish (Xiphophorus helleri) were exposed to polystyrene microplastics at different particle sizes (0, 1 μm, 5 μm, and 1∶1 mixture of them) and different concentrations (0, 1×106 particles·L-1, and 1×107 particles·L-1) in water environment for 72 h. The activities of digestive enzymes, antioxidant enzymes and immune enzymes were then determined. The results showed that compared with the control group, the activities of intestinal amylase, lipase and trypsin in all exposed groups were significantly increased (P < 0.05). The activities of superoxide dismutase and glutathione peroxidase decreased significantly (P < 0.05), while the activity of catalase increased significantly (P < 0.05). Moreover, the activities of malondialdehyde and lysozyme also increased significantly (P < 0.05). This study suggests that acute exposure to microplastics will interfere with food digestion and absorption of swordtail fish, and affect the activities of intestinal antioxidant enzymes and immune enzymes.

塑料制品因质轻、防水、绝缘、耐用和抗腐蚀等优点,在人类生产生活中得到广泛应用。据统计,全球每年的塑料使用量不少于3.5×108 t,塑料制品在自然环境下难降解,导致环境中的废弃塑料越来越多(侯淼淼等,2020)。废弃塑料如塑料袋、矿泉水瓶、一次性饭盒、废弃渔网等在物理、化学或其他因素作用下分解成更小的塑料碎片或颗粒,其中直径<5 mm的塑料碎片或颗粒被称为微塑料(Thompson et al., 2004)。部分微塑料颗粒会随降雨或地表径流等进入水环境,因其大小与浮游生物相似,而极易被水生生物误食,如:微塑料会引起牡蛎Crassostrea gigas、贻贝Mytilus edulis、鲱海鲷Pagellus erythrinus等消化道堵塞(Pascal & Patricia 2016Rossana et al., 2016Savoca et al., 2019),导致这些动物的摄食率降低,严重时甚至引起个体死亡等(Saskia et al., 2016)。此外,被水生生物误食的微塑料(Oliveira et al., 2013Wright et al., 2013Annika et al., 2016)还可以通过物理性损伤、载体效应(如富集其他污染物)、生物积累与食物链传递等途径产生一系列生态毒理效应,如:行为毒性(如运动能力下降、摄食能力降低)、生殖毒性(如生长发育缓慢、繁殖速度降低)、免疫毒性(如引发机体免疫炎症)、氧化应激(如造成机体氧化损伤)等(Moos et al., 2012Julia et al., 2014Rossana et al., 2016Chen et al., 2017)。Ali等(2017)将斑马鱼Danio rerio幼鱼暴露于聚苯乙烯微塑料环境中,其肠道过氧化氢酶(CAT)活性在暴露20 d时的表达量高于第10天;Wen等(2018)将七彩神仙鱼Symphsodon aequifasciata暴露于70~80 μm聚苯乙烯微塑料环境30 d后,发现其胰蛋白酶(TRY)活性显著降低;Tang等(2020)将泥蚶Tegillarca granosa暴露于30 μm微塑料环境中,发现其溶菌酶(LZM)活性随着微塑料浓度的升高而降低。因此,推测聚苯乙烯微塑料对剑尾鱼Xiphophorus helleri的消化、抗氧化及免疫酶活性也存在一定程度的影响,预期可能会造成消化酶活性降低、抗氧化酶活性和免疫酶活性升高。杨兵坤等:不同粒径微塑料对剑尾鱼肠道消化、抗氧化及免疫酶活性影响

剑尾鱼是一种小型热带淡水鱼类,也是我国首个通过审定的淡水鱼类实验动物,体型小、易饲养、繁殖力强、繁殖周期短、对多种重金属毒物较敏感,可作为水环境监测、水产药物安全性评价、化学用品毒性检测、动物疾病检验及生态毒理学研究的模式鱼类(吴淑勤等,2003)。关于微塑料对剑尾鱼的毒效研究较少,因此本文以剑尾鱼为实验对象,将其暴露于粒径1 μm、5 μm及混合粒径聚苯乙烯微塑料环境中72 h,测定肠道中消化酶、抗氧化酶和免疫酶活性,以期积累微塑料对鱼类生物毒性数据,并为深入研究微塑料的毒理学影响奠定基础。

1 材料与方法 1.1 实验材料

5月龄红眼红体雄性剑尾鱼购自陕西红剑尾鱼养殖基地,于实验室内曝气24 h的自来水中饲养与驯化。饲养与驯化过程中水体持续24 h曝气及活性炭处理,氧溶解度≥5.7 mg·L-1。驯化及实验过程中保持水温27 ℃±1 ℃,pH7.2~7.6,光周期14 h L∶ 10 h D。每天09∶ 00和17∶ 00喂食,饲料投喂量为剑尾鱼体质量的3%,饲料购于深圳市寸金观赏鱼饲料有限公司。聚苯乙烯微塑料为粒径1 μm与5 μm的球形单分散绿色荧光塑料微球,可均匀分散于水中,最大激发和发射波长为470 nm和526 nm,购于天津倍思乐色谱技术开发中心。

1.2 实验方法

在实验室驯化2周后,选取颜色和体型相近、活泼健康的个体(体质量1.63 g±0.29 g,体长5.6 cm±0.55 cm) 作为实验动物,浓度设置参考微塑料环境浓度调查结果及其对水生生物的毒性效应的研究方法(Watts et al., 2015Yin et al., 2018),试验共分为7组:M0组(无微塑料)、M1-1组(微塑料颗粒直径:1 μm,浓度:1×106粒/L)、M1-2组(微塑料颗粒直径:1 μm,浓度:1×107粒/L)、M2-1组(微塑料颗粒直径:5 μm,浓度:1×106粒/L)、M2-2组(微塑料颗粒直径:5 μm,浓度:1×107粒/L)、M3-1组(微塑料颗粒直径:1 μm与5 μm按颗粒数1∶ 1混合,浓度:1×106粒/L)和M3-2组(微塑料颗粒直径:1 μm与5 μm按颗粒数1∶ 1混合,浓度:1×107粒/L)。每个处理组设置3个重复,每个重复10尾,共210尾。暴露前配制各组聚苯乙烯微塑料溶液,并进行超声波处理,确保微塑料颗粒均匀分布于水环境中,暴露期间的环境与驯养时保持一致,依据《化学品鱼类急性毒性试验GB/T27861-2011》(中华人民共和国国家质量监督检验检疫总局,中国国家标准化管理委员会,2012)暴露72 h,实验进行至36 h时更换1/3提前曝气及活性炭处理过的水,并添加相应聚苯乙烯微塑料以维持暴露浓度。

72 h暴露实验结束后,立即使用MS-222麻醉剂麻醉,待鱼无反应后,用蒸馏水冲洗鱼身3次,冰上解剖。每个重复中取3尾鱼的肠道作为1个样本,每个处理组6个平行样本。剔除肠道外脂肪及内容物,用生理盐水冲洗后装入已编号的冻存管中,放入液氮中速冻,-80 ℃保存。取出冻存管中的肠道置于称量纸上准确称取其质量并记录,按照体积(mL)∶质量(g)=9∶ 1的比例加入预冷的生理盐水,随后在冰水浴中对肠道组织进行机械研磨,制成浓度为10%的组织匀浆,低温高速离心(3 000 r·min-1,15 min),取上清用于测定淀粉酶(AMS)、脂肪酶(LPS)、TRY、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH)、CAT、丙二醛(MDA)、LZM和总蛋白活性。所有试剂盒均购自南京建成生物工程研究所,测定方法及原理参照试剂盒说明书。

1.3 数据统计与分析

采用SPSS 20.0进行数据统计,运用单因素方差分析(One-Way ANOVA)分析对照组和暴露组的酶活性差异。数据以x±SD表示,显著水平设置为α=0.05。使用GraphPad Prism 8作图。

2 结果 2.1 1 μm聚苯乙烯微塑料对剑尾鱼肠道消化酶、抗氧化酶和免疫酶活性的影响

随着水环境中聚苯乙烯微塑料浓度的增加,AMS活性总体呈上升趋势:M0组的AMS活性为(0.404 5±0.004 0) U·mg-1,M1-1组为(0.417 1± 0.050 0) U·mg-1,未发生显著变化(P>0.05),M1-2组为(0.430 9±0.007 8) U·mg-1,升高显著(P<0.05)。LPS活性显著上升:M0组为(26.79±0.85) U·g-1,M1-1组和M1-2组显著上升[(35.90±1.30) U·g-1和(49.26±1.83) U·g-1P < 0.05]。TRY活性呈先上升后下降的趋势:M0组为(850.05±69.23) U·mg-1,M1-1组显著上升至(1 113.25±22.53) U·mg-1(P < 0.05),M1-2组显著下降为(969.67±16.66) U·mg-1(P < 0.05),但仍高于M0组(图 1)。

随着水环境中聚苯乙烯微塑料浓度的增加,SOD活性总体呈显著下降趋势:M0组为(122.46± 3.75) U · mg-1,M1-1组和M1-2组显著下降[(109.38± 2.08) U·mg-1和(101.83±2.68) U·mg-1P < 0.05]。GSH活性总体呈显著下降趋势:M0组为(83.08±3.19) U·g-1,M1-1组和M1-2组显著下降[(65.63±1.90) U·g-1和(72.53±2.47) U·g-1P < 0.05]。CAT活性总体呈上升趋势:M0组为(18.47±0.49) U·mg-1,M1-1组和M1-2组显著上升[(23.71±0.85) U·mg-1和(23.03±1.06) U·mg-1P < 0.05](图 1)。

随着水环境中聚苯乙烯微塑料浓度的增加,MDA活性总体呈上升趋势:M0组为(5.66± 0.11) nmol·mg-1,M1-1组和M1-2组显著上升[(6.84±0.13) nmol·mg-1和(6.70± 0.14) nmol·mg-1P < 0.05]。LZM活性呈显著上升趋势:M0组为(244.85±12.89) U·mL-1,M1-1组和M1-2组显著上升[(318.19±11.09) U·mL-1和(456.21±13.13) U·mL-1P < 0.05](图 1)。

2.2 5 μm聚苯乙烯微塑料对剑尾鱼肠道消化酶、抗氧化酶和免疫酶活性的影响

随着水环境中聚苯乙烯微塑料浓度的增加,AMS活性呈上升趋势:M0组为(0.404 5±0.004 0) U·mg-1,M2-1组为(0.436 5± 0.010 5) U·mg-1,升高显著(P<0.05),M2-2组为(0.505 8±0.005 6) U·mg-1,升高显著(P < 0.05)。LPS活性呈显著上升趋势:M0组为(26.79±0.85) U·g-1,M2-1组和M2-2组显著上升[(45.31±2.12) U·g-1和(50.13±1.79) U· g-1P < 0.05]。TRY活性呈先上升后下降的趋势:M0组为(850.05±69.23) U·mg-1,M2-1组显著上升为(1 094.60±82.97) U·mg-1(P < 0.05),M2-2组显著下降为(1 023.27±28.79) U·mg-1(P < 0.05),但仍高于M0组(图 2)。

随着水环境中聚苯乙烯微塑料浓度的增加,SOD活性总体呈下降趋势:M0组为(122.46±3.75) U·mg-1,M2-1组显著下降至(95.99±1.66) U·mg-1(P < 0.05),M2-2组显著下降至(101.40±2.31) U·mg-1(P < 0.05)。GSH活性总体呈下降趋势:M0组为(83.08±3.19) U·g-1,M2-1组和M2-2组显著下降[(57.74±0.70) U·g-1和(55.25±1.03) U·g-1P < 0.05]。CAT活性呈上升趋势:M0组为(18.47±0.49) U·mg-1,M2-1组和M2-2组显著上升[(21.01±0.66) U · mg-1和(23.08±0.61) U·mg-1P < 0.05](图 2)。

随着水环境中聚苯乙烯微塑料浓度的增加,MDA活性总体呈上升趋势:M0组为(5.66± 0.11) nmol · mg-1,M2-1组和M2-2组显著上升[(6.83± 0.90) nmol · mg-1和(6.82±0.71) nmol·mg-1P < 0.05]。LZM活性呈显著上升趋势:M0组为(244.85±12.89) U·mL-1,M2-1组和M2-2组显著上升[(359.55±6.44) U·mL-1和(476.67± 9.14) U·mL-1P < 0.05](图 2)。

2.3 混合粒径聚苯乙烯微塑料对剑尾鱼肠道消化酶、抗氧化酶和免疫酶活性的影响

随着水环境中聚苯乙烯微塑料浓度的增加,AMS活性总体呈上升趋势:M0组为(0.404 5±0.004 0) U·mg-1,M3-1组和M3-2组上升[(0.441 2±0.008 9) U·mg-1和(0.457 4±0.009 5) U·mg-1P < 0.05]。LPS活性呈先上升后下降的趋势:M0组为(26.79±0.85) U·g-1,M3-1组显著上升至(47.45±1.37) U·g-1 (P < 0.05),M3-2组下降至(53.93±1.24) U·g-1,仍显著高于M0组(P < 0.05)。TRY活性呈上升趋势:M0组为(850.05±69.23) U·mg-1,M3-1组显著上升至(1 196.24±114.03) U·mg-1(P < 0.05),M3-2组显著上升至(1 020.18±45.34) U·mg-1(P < 0.05),但仍高于M0组(图 3)。

随着水环境中聚苯乙烯微塑料浓度的增加,SOD活性总体呈下降趋势:M0组为(122.46±3.75) U·mg-1,M3-1组显著下降至(105.53±3.08) U·mg-1(P < 0.05),M3-2组下降至(109.01±1.68) U·mg-1(P < 0.05)。GSH活性总体呈下降趋势:M0组为(83.08±3.19) U·g-1,M3-1组和M3-2组显著下降[(72.83±0.73) U·g-1和(69.67±0.44) U·g-1P < 0.05]。CAT活性呈上升趋势:M0组为(18.47±0.49) U · mg-1,M3-1组和M3-2组显著上升[(24.28±0.73) U·mg-1和(24.08±0.35) U·mg-1P < 0.05](图 3)。

随着水环境中聚苯乙烯微塑料浓度的增加,MDA活性呈上升趋势:M0组为(5.66±0.11) nmol·mg-1,M3-1组和M3-2组显著上升[(6.51±0.94) nmol · mg-1和(6.56±0.12) nmol · mg-1P < 0.05]。LZM活性呈上升趋势:M0组为(244.85±12.89) U·mL-1,M3-1组和M3-2组显著上升[(354.12±9.02) U·mL-1和(353.51±8.29) U·mL-1P < 0.05](图 3)。

3 讨论 3.1 不同粒径微塑料对剑尾鱼肠道消化酶活性的影响

在水生生物中,已经证实消化速率会影响机体的消化吸收能力,而消化酶活性水平是反映机体消化速率和能量吸收的相关指标(Gu et al., 2019)。其中, AMS、LPS和TRY分别参与碳水化合物、脂质纤维素和蛋白质的消化过程(Charlene et al., 2021),在消化吸收过程中扮演重要角色。本研究发现摄入不同粒径微塑料后剑尾鱼肠道消化酶活性均发生改变,1 μm粒径组、5 μm粒径组和混合粒径组中AMS、TRY和LPS 3种消化酶活性的变化结果基本一致,其中5 μm粒径组较其他暴露组对AMS活性影响更大,但差异不显著,各粒径组暴露结果显示,低浓度组中TRY活性升高显著,高浓度组中AMS和LPS活性升高显著,推测可能是微塑料暴露粒径越大、浓度越高对剑尾鱼肠道消化吸收能力影响更显著。已有研究表明,将银鱼Prolosalanx hylocranius幼鱼暴露于粒径<40 μm聚氯乙烯微粒的环境96 h后,其肠道TRY活性显著升高(Romano et al., 2018),机体的消化吸收能力降低,与本研究结果相似。而Gu等(2020)将大黄鱼Pseudosciaena crocea暴露于粒径100 nm、浓度1.8×1013粒/mL的聚苯乙烯微塑料环境中14 d后,与对照组相比,其肠道TRY活性显著降低,但AMS和LPS活性没有显著变化,结果仍会引起机体消化系统紊乱。Huang等(2020)将孔雀鱼Poecilia reticulata暴露于粒径40 μm聚苯乙烯微塑料环境中28 d后,其肠道AMS、TRY和LPS活性均降低,导致机体从食物中获得能量储备的能力下降。Wang等(2020)将贻贝在粒径2 μm聚苯乙烯微球环境中暴露14 d后,与对照组相比,其AMS和LPS活性均被显著抑制,导致机体消化能力下降。由此看来,无论消化酶活性是升高还是降低,不同粒径和浓度微塑料暴露均会对机体消化吸收造成影响,而本研究中消化酶活性升高的原因可能是剑尾鱼个体较小、肠腔狭窄,摄入的微塑料在消化道内聚团、堆积造成机械阻塞,产生虚假“饱腹感”,导致机体摄入营养和能量不足(Watts et al., 2015)。机体通过增加消化酶活性的方式来提高对食物的消化率,补偿其能量摄入的不足,从而维持机体正常需要。但微塑料对消化吸收的影响仅用消化酶活性的变化无法系统阐释,接下来会进行更多的研究,如剑尾鱼肠道组织切片及肠道微生物菌群分析等。

3.2 不同粒径微塑料对剑尾鱼肠道抗氧化酶活性的影响

机体内主要的抗氧化酶SOD、GSH和CAT等在抑制和分解自由基和预防氧化损伤等方面发挥着关键作用(Li LL et al., 2020)。已有研究表明,微塑料可以引起珊瑚Anthozoan、斑马鱼、贻贝、南美白对虾Litopenaeus vannamei、金鱼Carassius auratus等抗氧化酶活性的改变,从而诱发机体氧化应激(Soares et al., 2020Wang et al., 2020Hsieh et al., 2021Xu et al., 2021)。本实验中,5 μm粒径组较其他暴露组对SOD和GSH活性影响更显著,可能是该组剑尾鱼肠道氧化损伤更严重。各暴露组中SOD和GSH活性显著降低,CAT活性显著升高,可能由于SOD和GSH是保护细胞免受氧化应激的内源性抗氧化酶,具有协同作用,同时CAT活性增加以维持体内抗氧化过程的动态平衡。Lei等(2018)将斑马鱼幼鱼暴露于粒径0.1 μm、浓度1.5×1013粒/L聚苯乙烯微塑料环境中,其GSH活性下降,推测微塑料暴露可能会导致机体能量代谢紊乱,诱发氧化损伤。Tang等(2018)将石珊瑚Pocillopora damicornis暴露于粒径1 μm、浓度9×1010粒/L聚苯乙烯微塑料环境中12 h后,发现其SOD活性升高,机体出现氧化应激。而Carlo等(2015)将紫贻贝Mytilus galloprovincialis暴露于粒径2 μm聚苯乙烯微塑料环境中,其SOD活性降低,且机体内脂质出现过氧化损伤,这与本研究结果基本相似,抗氧化酶活性的改变表明微塑料的摄入确实会干扰到机体内抗氧化系统稳态,当面对环境中微塑料的胁迫时,机体内抗氧化应激系统启动,以抵御不良环境的刺激。但是微塑料暴露在肠道组织中诱导氧化损伤的详细机制还需要进一步研究。

3.3 不同粒径微塑料对剑尾鱼肠道免疫酶活性的影响

MDA和LZM在非特异性免疫中起着至关重要的作用,被认为是评价机体内免疫状况的重要指标。MDA是机体内重要的氧自由基的代谢产物,可间接反映细胞损伤程度(Xia et al., 2020)。而LZM由中性粒细胞和巨噬细胞产生,通过激活补体和巨噬细胞来清除病原菌(Liu et al., 2019)。在本研究中,5 μm粒径组MDA和LZM的活性较其他2组升高更显著,可能是该组剑尾鱼肠道免疫损伤更严重,但各粒径组中MDA的结果未出现浓度效应,表明在环境污染或不利条件下,生物体试图通过免疫反应来适应相关的不良环境。淡水微藻Chlamydomonas reinhardtii在粒径300 nm、浓度100 mg·L-1微塑料环境中,MDA活性显著升高,并出现脂质损伤(Li SX et al., 2020),与本研究结果相似。大型蚤Daphnia magna暴露于粒径1 μm、浓度0.1 mg·L-1的聚苯乙烯微塑料环境中48 h后,MDA活性升高,并引起组织损伤(Zhang et al., 2019)。鲤鱼Cyprinus carpio暴露于粒径6 μm、浓度50 mg·L-1微塑料环境中60 d后,MDA活性显著降低,相关基因表达量减少(Xia et al., 2020)。而将中华绒螯蟹Eriocheir sinensis暴露于粒径5 μm、浓度40 mg·L-1的微塑料环境中7 d、14 d、21 d后,LZM活性先升高后降低,出现酶活性被抑制的现象并引起毒性效应(Liu et al., 2019)。本研究采用的72 h急性微塑料暴露实验中,尚未观察到免疫酶活性先升高后降低的趋势,下一步计划采用短期和长期结合暴露实验,观察是否会出现免疫酶活性先升高再降低的趋势,并结合个体、组织、细胞和基因水平进行测试,以便为进一步研究微塑料的免疫毒性效应奠定基础。

在本研究中,5 μm粒径组较其他暴露组对剑尾鱼肠道消化酶、抗氧化酶及免疫酶活性影响更大,但差异不显著。不同粒径聚苯乙烯微塑料均会影响消化酶、抗氧化酶及免疫酶活性的表达,以此干扰剑尾鱼对食物的消化和能量的吸收,并对肠道的抗氧化能力和免疫能力造成影响。
