摘要[目的]对纳米通道中离子电流调制机制的理解,是发展纳米农业检测传感器的关键。[方法]采用分子动力学模拟的方法,研究受限纳米通道中单壁碳纳米管的荷电量对离子电流阻塞的影响。[结果]仿真结果表明:电中性的单壁碳纳米管进入通道中时产生的阻塞电流始终小于基准电流。在碳纳米管带电荷且荷电量增加的情况下,当溶液浓度较低时,阻塞电流大于基准电流;当溶液浓度较高时,阻塞电流小于基准电流,出现电流交叉现象。[结论]单壁碳纳米管过孔时其体积占位对电渗流的影响以及其本身荷电量对纳米通道中离子浓度的影响相互竞争,共同决定着纳米通道中的离子电流。

Mechanism of the impact of single-walled carbon nanotubes on ionic current in a nano-channel

Abstract: [Objectives] Understanding the mechanism of ionic current modulation in nano-channels is the key to the development of nano agricultural detection sensors. [Methods] Molecular dynamics simulation was used to study the impact of charge density of single-walled carbon nanotubes on the ionic current blockades in a confined nano-channel. [Results] The simulation results indicated that when an electrically neutral single-walled carbon nanotubes entered the nano-channel, the blockade current was always less than the reference current. With increasing charge number on the carbon nanotube, and at lower solution concentrations, the blockade current was greater than the reference current. When the solution concentration was high, the blockade current was less than the reference current, and the phenomenon of current crossover occurred. [Conclusions] The effect of the volume occupied by the single-walled carbon nanotubes on electroosmosis and the effect of their own charge on the ion concentration in the nano-channel compete with each other, and together determine the ionic current in the nano-channel.

科学技术的进步带动着现代农业朝着精细化、信息化的方向发展。纳流控作为新兴科技之一, 在农业领域有着巨大的应用潜力, 其可用于实现植物病害的预知以及农产品中生物、化学污染残留的检测等, 大大改善了农业生产的现状[1-4]。纳米孔作为一种高通量的检测传感器, 已实现了对大量物质的检测, 如植物病毒[5]、蛋白质[6-8]、纳米颗粒[9-10]、金属离子[11]和DNA[12-16]等。其检测的原理是将待测物的信息以电流信号的形式输出, 通过监测纳米通道中待测物运输引起的离子电流变化来分析和检测待测物的相关信息。在利用外加电场来驱动待测物通过纳米通道时, 待测物改变了通道内的电阻, 会引起离子电流发生变化 , 即电阻脉冲法。因其在农业、生命科学等各领域的物质检测方面有着巨大的应用潜力而得到了广泛的关注 。但一些研究表明, 待测物在通过纳米通道时并不只是单纯地使电流下降, 如Wu等[5]发现刚性杆状的烟草花叶病毒在通过纳米孔时, 其旋转会影响电流信号强度; Qiu等[10]研究的不同直径、带有不同电荷密度的纳米颗粒通过纳米通道时也产生了类似的电流现象, 同时也表明纳米通道内电荷之间复杂的相互作用并不能用已有的简单模型进行描述; Zhang等[12]发现在不同直径的纳米孔内离子化合价的不同会使DNA在孔内输运也不同, 电流现象也因此产生差异; Lv等[14]发现在不同直径和层数的石墨烯纳米孔中DNA自由能的变化会对其运输产生影响, 从而改变纳米孔的电导率; Zanjani等[21]研究发现纳米杆的体积占位、纳米通道的壁面电荷以及纳米杆自身所带电荷三者的共同作用使纳米杆通过不同直径的纳米通道时会产生或增大或减小的电流现象。

目前, 大多数研究集中于待测物在纳米通道内运输时对调制电流的影响, 而对待测物通过纳米通道时对调制电流影响机制的研究较少。对受限纳米通道中壁面电荷、溶液浓度以及待测物荷电量之间复杂的相互作用的理解比较缺乏。本文通过分子动力学模拟的方法, 采用单壁碳纳米管(single-walled carbon nanotubes, SWCNT)分子模型, 对不同溶液浓度下, 荷电量不同的单壁碳纳米管分子通过纳米通道时, 通道中粒子的速度及离子的径向密度分布等进行统计研究, 详细阐述纳米通道内离子电流调制的机制, 为纳米农业检测传感器的开发提供重要参考。

1 分子动力学模型

图 1为利用GROMACS软件包建立的圆柱形纳米通道流体运输模型的截面示意图。整个模拟系统沿着Z方向依次分为左液池、圆柱形纳米通道和右液池3个部分, 其三维尺寸为5.5 nm×5.5 nm×10 nm。圆柱形纳米通道建立在三维尺寸为5.43 nm×5.43 nm×5.7 nm的长方体单质硅上, 其直径为3.1 nm, 中心位于长方体几何中心处。纳米通道位于整个模拟系统的中心位置, 其内壁上均布有一定数量的离散元电荷, 电荷密度为-1 e·nm-2。单壁碳纳米管的直径为0.31 nm, 长度为2 nm, 其表面电荷密度分别为0、+1.299、+2.598 e·nm-2。溶剂选用SPC(simple point charge)水分子模型, 初始时体系中含有5 362个水分子。溶液浓度为0.3、0.6、0.9、1.2、1.5、1.8 mol·L-1。为维持体系的电中性, 钠离子及氯离子的数量由溶液的浓度、通道的壁面电荷和碳纳米管的荷电量来共同确定[22]

模拟选用GROMACS力场, 采用Lennard-Jones和库伦作用势对体系中粒子间的相互作用进行描述。范德华作用的处理采用势能截断法, 截断半径取1.108 nm[23], 库伦静电作用的处理采用Particle-Mesh Ewald法。选择Leap-frog算法对运动微分方程进行求解, 积分步长为2 fs[24]。模拟选择等温等压系统, 并运用Berebdsen热浴法和Berebdsen压浴法对体系的温度和压强进行耦合以维持体系的温度在298 K, 压强为1个标准大气压, 耦合时间常数为0.1 ps[25]。整个模拟过程中, 沿模拟体系的Z轴正方向施加电场强度为1 V·nm-1的外加电场。模拟初始, 各粒子的初始速度依据Maxwell分布进行赋值, 初始位置随机分布于模拟体系内。在整个模拟过程中, 固定通道最外层硅原子不动, 以利于碳纳米管在通道中的运输以及通道内、外离子的交换, 使得整个模拟更加符合工程实际。先对模拟体系进行4 ns的预处理, 使其达到相对平衡的状态, 再对其施加电场模拟4 ns来达到新的平衡。对后4 ns模拟中产生的所需要的数据进行统计分析。

2 分子动力学模拟结果及分析

在分子动力学模拟过程中, 基准电流被定义为离子在不含单壁碳纳米管的纳米通道中定向移动而产生的电流; 而把单壁碳纳米管在纳米通道中运输时离子和单壁碳纳米管移动而产生的电流称作阻塞电流。图 2为通道壁面电荷密度为-1 e·nm-2情况下, 通道内无碳纳米管及含有荷电量不同的碳纳米管时, 在外加电场力的作用下对离子电流的统计。

图 2可知:在相同溶液浓度下, 电中性的单壁碳纳米管进入纳米通道时产生的阻塞电流始终小于基准电流。在溶液浓度较低时, 荷电的单壁碳纳米管分子进入纳米通道中产生的阻塞电流大于基准电流; 但随着溶液浓度的增大两者的差距会逐渐缩小, 最终在达到某一浓度时阻塞电流小于基准电流, 这种荷电单壁碳纳米管所引起的纳米通道内离子电流交叉的现象与文献[13]所报道的现象相类似。此外, 在相同的溶液浓度下, 单壁碳纳米管的荷电量越高, 其产生的阻塞电流就越大。

为解释上述现象, 图 3对纳米通道内电渗流的速度与钠离子、氯离子的轴向平均速度进行统计。对比可见:离子的速度随溶液浓度的增大而减小, 且在相同的溶液浓度下, 不论单壁碳纳米管是否带电荷, 其在纳米通道中时, 离子的速度均低于不含单壁碳纳米管时的速度。这是由于纳米量级尺度下流体的黏度与流体的局部密度呈正相关性[26], 密度越大则流体的黏度越大。在外加电场的作用下, 纳米通道中电渗流的速度(V)如式(1)所示:

式中:E为外加电场强度; ε0为真空介电常数; εr为溶液相对介电常数; ζzeta电势; η为溶液的黏度。

由公式(1)可知:保持外加电场强度不变, 电渗流的速度与溶液的黏度成反比, 纳米通道内流体的密度越大黏度越高, 电渗流的速度越小。因此单壁碳纳米管进入通道后, 由于其体积占位作用, 纳米通道内流体的局部密度增大, 使得溶液黏度加大而导致离子速度降低引起离子电流减小。另外, 由图 3可以看出:相同溶液浓度下, 当带电荷的单壁碳纳米管进入纳米通道时, 通道中各粒子的速度比中性单壁碳纳米管进入时大, 且其荷电量越大, 粒子速度越大。这是因为随着单壁碳纳米管的荷电量增加, 其所受的电场力增大, 速度增大。荷电的单壁碳纳米管的运动方向与电渗流方向一致, 相比于中性的单壁碳纳米管, 其对电渗流的“阻碍”作用降低, 使得电渗流的速度相对增大。电渗流速度的增大使得其对钠离子的“阻碍”作用减小, 钠离子的速度相对增大。荷正电的单壁碳纳米管进入通道的同时会吸引氯离子进入通道中心区域, 该区域氯离子所受通道壁面的约束作用较弱, 速度较大, 使得氯离子的平均速度相对增大。

图 4是溶液浓度为0.9 mol·L-1时, 纳米通道内钠离子和氯离子的径向密度分布图。可以看出, 无论通道中有无单壁碳纳米管或其是否带电荷, 钠离子在纳米通道内的径向密度分布基本相同。当中性的单壁碳纳米管进入纳米通道时, 通道内氯离子的径向密度分布与不含单壁碳纳米管的通道中的密度分布相近; 当带电荷的单壁碳纳米管进入纳米通道时, 氯离子的密度会比不含单壁碳纳米管的通道中的密度大, 且单壁碳纳米管的荷电量越大, 氯离子的密度越大。

为进一步解释单壁碳纳米管对离子浓度变化的影响, 统计了溶液浓度为0.9 mol·L-1时, 带有不同电荷的单壁碳纳米管进入纳米通道后离子距离单壁碳纳米管中心的径向密度分布情况。从图 5可以看出:带有不同正电荷密度的单壁碳纳米管自由运动时, 钠离子距其中心的径向密度分布基本相似; 而氯离子距单壁碳纳米管中心的径向密度则明显不同, 单壁碳纳米管附近的氯离子的径向密度会随其电量的增加而增大, 对此可认为是静电作用使带正电的单壁碳纳米管会吸引氯离子而不吸引钠离子所致。因此, 当带电荷的单壁碳纳米管进入纳米通道时会吸引氯离子同时进入纳米通道, 引起通道中离子浓度的增加。

通过上述分析可知:当电中性的单壁碳纳米管进入纳米通道时, 其对通道中离子的浓度基本没有影响, 但单壁碳纳米管的体积占位会影响通道内电渗流的形成, 引起钠、氯离子的轴向速度减小, 使阻塞电流始终小于基准电流; 当溶液浓度较低时, 带电荷的单壁碳纳米管进入纳米通道后, 随其荷电量的增加, 吸引至通道内维持通道电中性的反离子数目增多, 且在电场作用下其在纳米通道内的轴向移动速度增大, 对离子运动的阻碍作用会减弱, 引起离子的轴向运动速度相对增大, 使阻塞电流大于基准电流; 当溶液浓度较高时, 反离子数量的变化对通道中离子浓度的影响相对较弱, 如图 6所示, 单壁碳纳米管的体积占位对离子速度的影响大于其荷电对离子浓度的影响, 因此阻塞电流会小于基准电流。

3 结论

采用分子动力学仿真的方法研究了在不同溶液浓度下, 荷电量不同的单壁碳纳米管通过受限纳米通道时对通道内离子电流的影响。研究结果表明:当电中性的单壁碳纳米管进入纳米通道时, 通道中反离子的浓度不发生变化, 其体积占位影响电渗流的形成, 使通道内离子的轴向速度下降, 导致阻塞电流小于基准电流。在溶液浓度较低的情况下, 带电荷的单壁碳纳米管进入纳米通道时, 随其荷电量的增加, 通道中心区域会出现更多的反离子来维持通道的电中性, 且在外加电场力的作用下, 单壁碳纳米管的轴向速度增大, 引起离子轴向运动速度相对增大, 使阻塞电流大于基准电流。在溶液浓度较高的情况下, 带电的单壁碳纳米管进入纳米通道时, 其荷电量对通道中离子浓度的影响相对较小, 其体积占位对电渗流的影响占主导地位, 使阻塞电流最终小于基准电流。综上可知:单壁碳纳米管过孔时其体积占位对电渗流的影响以及其本身荷电量对纳米通道中离子浓度的影响相互竞争, 使纳米通道中阻塞电流与基准电流之间随溶液浓度变化产生交叉现象。本研究对纳米农业检测传感器的开发提供参考, 推动纳流控技术在现代农业领域中的应用。
