双酚A (bisphenol A,BPA)是一种合成化合物,作为聚合物单体,广泛应用于合成聚碳酸酯、环氧树脂、聚砜类和聚酯树脂等。聚碳酸酯主要用于生产食品容器和包装材料,如奶瓶、餐具、微波用容器和塑料瓶等。环氧树脂常见于存储食品的金属罐和瓶盖的涂层,加热这些材料会导致BPA发生迁移至其接触的食品。此外,BPA在生活中随处可见,如热敏纸、粘合剂、医疗设备、画笔以及太阳镜等。粗略估计,每年大约有100 t的BPA被释放进入环境当中[1]。环境中的BPA以多种方式进入食物链,根据欧洲食品安全局2014年估算,每人每日能够承受的BPA摄入量为每公斤体重0.005 mg。在欧盟和中国,食品接触材料的BPA迁移量不允许超过0.6 mg/L,日本则是2.5 mg/kg。BPA在结构上类似雌激素,会干扰正常的生理过程,被认定为内分泌干扰物[2]。即使是皮克量级的BPA含量,也会对甲状腺、大脑、外分泌胰腺、卵巢、生殖器和免疫系统造成不良影响。研究还表明BPA与肥胖、糖尿病、心血管疾病、肝毒性、神经毒性以及行为问题有关系[3-5]。因此,研究环境中BPA的检测分析方法具有重要意义。
本文主要综述环境和人体中BPA的前处理技术,分析比较了常用检测方法的优缺点,也介绍了如传感器检测法等新的方法,并结合目前的研究进展对该领域今后的研究工作做了展望。
1 BPA前处理方法环境和人体样品中BPA的含量通常很低,甚至为痕量级别,而且基质中存在严重的离子抑制或者增强现象,因此,对样品净化减少样品中基质的干扰效应和富集BPA的前处理过程是分析检测BPA的关键环节[6]。
1.1 液液萃取(liquid-liquid extraction,LLE)LLE是非常传统的前处理方式,操作简单、有效且价格低廉。缺点是有机试剂消耗量大,不是环境友好型的前处理方式。BPA的萃取溶剂通常包括乙腈、甲醇、1-氯丁烷和正己烷。通过结合微萃取技术、绿色化学技术和其他加速过程的方法,既可以提高萃取效率,又可以减少萃取所需的溶剂量。浊点萃取技术(cloud point extraction,CPE)作为一种绿色的LLE方法,以表面活性剂溶液的增溶和分相来实现物质的富集和分离,应用于水样中BPA和萘酚的萃取[7];该方法的有机溶剂消耗量少,只需200 μL浓度为0.1%的Triton X-114。完成萃取操作后,表面活性剂层用200 μL乙腈稀释,之后进行毛细管电泳,结合紫外检测器进行测定。该方法测定BPA的定量限为1.67 g/L。CPE的缺点在于相分离的速度限制了能够处理的样品量。萃取技术的微型化已经成为绿色化学的趋势。液相微萃取(liquid-phase microextraction,LPME)和分散液液微萃取(dispersive liquid-liquid microextraction,DLLME)是新兴的微萃取技术。LPME的优点在于操作简便、萃取时间短和高灵敏度,缺点在于精确度不足。Wu等[8]报道了一种柱前衍生化结合DLLME方法,用于同时测定六种类固醇和内分泌干扰素(endocrine disrupting chemicals, EDCs),包括BPA。结合液相色谱-荧光检测器,所有化合物的检出限均在(0.02~0.07) μg/L范围内。Liu等[9]比较了正相分散液液微萃取方法和反相分散液液微萃取(reversed-phase dispersive liquid-liquid microextraction,RP-DLLME)方法在食用油中BPA的含量分析,结果显示RP- DLLME方法的检测效果更好,其检出限为2.5 ng/g,实际样品的回收率为89.5%~99.7%。
1.2 微波辅助萃取技术(microwave-assisted extraction,MAE)MAE是将微波技术与萃取技术相结合,在样品前处理阶段借助微波进行萃取,之后结合固相萃取SPE进一步除杂纯化,再用仪器进行分析。MAE已经成功地应用于BPA的预浓缩和提取中。Pedersen等[10]将冰冻幼虹鳟的肌肉和肝脏置于充满液氮的研钵内敲碎,取1 g样品加入内标,用20 mL二氯甲烷:甲醇(2:1)溶液进行微波辅助萃取,过滤,加入0.9%氯化钾溶液,离心,弃去二氯甲烷层,旋干,用1 mL甲醇:己烷(1:20)溶解。之后用500 mg Sep-Pak NH2柱进行固相萃取。该技术与液相色谱—质谱技术相结合,具有溶剂消耗量小(20 mL),耗时短(25 min),样品量小(1 g新鲜组织),定量限低(肌肉:10 ng/g;肝脏:50 ng/g)的优点。MAE与气相色谱—质谱技术结合[11],连续快速测定河流泥沙中的EDCs(包括BPA),采用甲醇为萃取溶剂,在110 ℃下萃取15 min,回收率均大于74%。快速微波辅助离子液体微萃取(microwave-assisted ionic liquid microextraction)也成功应用于萃取牛奶中的BPA和四溴双酚A[12]。该方法操作仅需5 min,检出限为(0.01~0.02) μg/L,回收率为78.2%~99.8%。与传统的LLE方法相比,MAE具有操作简便、萃取效率高、耗时少、回收率高、成本低以及溶剂消耗量少等诸多优点,也更符合绿色化学的环保趋势。
1.3 固相萃取技术(solid-phase extraction, SPE)SPE由于操作简便且纯化效果良好,是目前最常用的前处理方法[13-14],该技术在BPA检测方面的应用已有比较完整的综述[15]。Asimakopoulos等[16]将混合固相萃取—预沉淀技术与LC-MS/MS技术相结合,应用于萃取纯化血液中的EDCs。该方法的仪器检出限为0.2 ng/mL。固相微萃取技术(solid-phase micro-extraction,SPME)是在SPE基础上发展出的更环保的萃取技术。Jiao等[17]用异烟酸铜作为吸附剂进行SPME,用HPLC进行分析检测大气颗粒物中的BPA及其他EDCs。该方法检出限为(2.0~8.5) ng/L,回收率为81.2%~119.2%。电子加强固相微萃取(electro-enhanced solid-phase microextraction,EESPME)方法[18]采用商业化的SPME纤维,萃取纯化血液和海水中的邻苯二甲酸酯和BPA。结合GC-MS测定,检出限为(0.004~0.15)μg/L。Zhang等[19]用三维石墨烯涂层纤维作为工作电极,进行EESPME萃取BPA,和传统SPME相比,萃取效率提高了3.2倍。和气相色谱结合,该方法在(0.1~10)μg/mL范围有良好线性(R2=0.993 5),检出限是0.006 μg/mL。应用于3种不同的热敏纸检测,检出范围在(0.696~3.78) mg/g。磁固相萃取(magnetic solid phase extraction,MSPE)以磁性/可磁性材料作为吸附剂,在磁场作用下分离目标物[20]。Du等[21]用磁性氨基功能化微孔有机网络复合材料进行MSPE,测定水样、饮料以及果汁中的BPA等EDCs,回收率为80.3%~109.5%,检出限为(0.015~0.030)μg/L。SPE的主要缺点是缺乏特异性[22],对样品的富集倍数有限,对检测器的灵敏度要求高,且价格比较昂贵。
1.4 分子印迹固相萃取技术(molecularly imprinted solid-phase extraction, MIP-SPE)分子印迹聚合物(molecularly imprinted polymer,MIP)是为分析物量身定制的材料,能够特异性识别指定分析物,已经被广泛应用于选择性吸收和检测各类化合物。很多BPA-MIPs已经被成功应用于不同样品中BPA的萃取和测定。分子印迹固相萃取技术的缺点为富水体系选择性不高、制备过程模板分子渗漏、洗脱困难和吸附量不够理想等。有研究用不同的印迹分子制备了系列BPA-MIPs,富集和检测样品中的BPA[23]。将甲基丙烯酰丙基三甲基氯化铵(3-acrylamido-N, N, N-trimethylpropan-1-ammonium chloride)作为带正电的功能单体,制备了1种亲水性的BPA-MIP,已成功用于多种含水试样的直接测定,并无需进行前处理[23]。新型多孔式空心虚拟分子印迹材料被报道应用于BPA的吸附与识别,采用TBBPA作为模板分子,中孔MCM-48纳米微球作为载体[24]。该材料具有吸附BPA容量大、速率高等优点,与HPLC相连,用于检测自来水中BPA含量,该方法的检出限为3 ng/mL,回收率为95%~105%。磁性分子印迹聚合物(magnetic molecularly imprinted polymers,MMIPs)为新型印记技术,同时包含了表面印记技术和磁载体的优点。Zhang等[25]用碳纳米管作为载体,制备了一种新型MMIP(MWNT@MMIPs),用BPA作为模板分子,甲基丙烯酸和环糊精作为二元功能单体,乙二醇二甲基丙烯酸酯作为连接剂,能够特异性识别BPA,对BPA的最大吸附量为49.26 μmol/g。通过HPLC进行分析检测,该材料成功吸附提取自来水、雨水和湖水中的BPA,回收率分别为89.8%~95.4%、89.9%~93.4%和87.3%~ 94.1%。MIP-SPME技术,是将MIP和SPME相结合,用于复杂环境样品的前处理,测量痕量有机污染物含量。由于该方法选择吸附能力强以及适用性广泛,目前已成为一个研究热点。BPA-MIP包裹的SPME纤维已经被成功应用于环境样品中BPA的检测。该方法和HPLC结合,检出限为(2.4~38.9) ng/mL,回收率为81.6%~92.5 % [26]。Xie等[27]将多模板分子印迹聚合物包裹于介孔二氧化硅包覆的磁性氧化石墨烯表面,制备了磁性多模板分子印迹聚合物,将其应用于检测水样中的BPA、4-叔辛基苯酚和4-正壬基酚,表现出良好的吸附选择性,且该材料可以重复使用。绿色分析化学是分析化学发展的一个趋势,主要目的是尽最大可能降低环境污染。样品前处理过程是分析方法学的重要组成部分,由于对有机溶剂的消耗量大,最有可能对环境造成污染。目前比较新的样品处理方法,诸如浊点萃取技术CPE、液相微萃取LPME、微波辅助萃取技术MAE、固相微萃取技术SPME以及分散液液微萃取DLLME应用在不同基质中检测BPA,正是应运而生的绿色处理手段。其中,SPME是最受欢迎且应用最多的前处理方法。为了增加分离选择性,高选择性材料诸如MIP就作为光纤涂层而应用于SPME。
2 BPA检测分析方法环境样品中的BPA浓度低且样品基质非常复杂,有效的前处理净化过程之后所采用的检测分析手段同样重要。
2.1 液相色谱法(liquid chromatography,LC)和液相色谱—质谱法(liquid chromatography-mass spectrometry,LC-MS)液相色谱法是最常用的BPA检测方法,质谱MS作为检测器,具有高特异性和高灵敏度的特点,LC-MS和LC-MS/MS是目前定量检测BPA最广泛应用的技术。Zhuang等[28]通过重氮化偶联反应将BPA衍生化,形成偶氮化合物,在410 nm处有最强的吸收强度,结合LC-UV进行检测,回收率为93.6%~99.3%,检出限为0.15 μg/mL。Yazdinezhad等[29]用磁性分离的前处理技术结合LC-MS/MS分离和检测软饮中的BPA。该方法定量限为0.03 ng/mL,低于通常报道的软饮料中的BPA含量值。ter Halle等[30]用多孔有机凝胶材料结合LC-MS/MS检测尿样中的BPA,该方法快速、环保。在选择的最佳条件下,尿样中的回收率为80%,检出限为5 μg/L。Zhou等[31]用温控离子液体分散液相微萃取LPME测定了水样中的BPA,正壬基酚和叔丁基苯酚。该研究中,用1-辛基-3-甲基咪唑六氟磷酸盐(1-octyl-3-methylimidazolium hexafluorophosphate)作为萃取剂。采用液相色谱法荧光检测器(LC-FLD)进行检测,检出限为(0.23~0.48) μg/L。另外,Zhou等[32]建立了一种敏感度高,选择性强的在线SPE-LC同位素稀释质谱法,用于检测尿样中的BPA、双酚F、双酚S和其他11种酚类。该方法完全自动化,而且只需要100 μL的样品,BPA的检出限为0.1 ng/mL。Kolatorova等[33]提出了同时测定血浆中的双酚类、羟基苯甲酸酯类和雌激素类的LC-MS/MS方法,该方法采用了丹酰氯衍生化来提高敏感度,BPA的检出限为41.6 pg/mL。
2.2 气相色谱-质谱法(gas chromatography-mass spectrometry,GC-MS)GC-MS检测BPA及其他内分泌干扰物。由于BPA挥发性弱,在进行GC-MS分析之前,需要进行衍生化来增加挥发性,以提高峰强度和分辨率。最常用的衍生剂为N, O-双(三甲基硅基)三氟乙酰胺(BSTFA)、BSTFA+1%三甲基氯硅烷(TMCS)和N-甲基-N-(三甲基硅基)三氟乙酰胺(MSTFA)。Ros等[34]对比了GC-MS和LC-MS/MS两种方法。GC-MS方法流程为:样品制备采用聚醚砜作萃取剂,用乙酸乙酯或甲醇进行复溶,用BSTFA+1% TMCS进行衍生化,之后用GC-MS进行检测分析。LC-MS/MS方法不需要衍生化,样品制备时间短,BPA检出限更低:(2~63)ng/L,但该方法不能检测低浓度的壬基酚类化合物。GC-MS和LC-MS/MS相比,能够同时测定的化合物更多,且重复性好,准确度高,但由于GC-MS需要进行比较繁琐的样品制备和清洗程序,以及衍生化的步骤,使得它在大批量样本的检测应用中受到了限制。
2.3 毛细管电泳(capillary electrophoresis,CE)CE和其他的分析技术相比,具有分辨率高、试剂及样品消耗少等优点。因此广泛应用于BPA及其类似物的检测中。Mei等[35]制备了一种BPA-分子印迹聚合物微球,将其作为SPE吸附剂,用于BPA的萃取。和C18 SPE相比,该材料对BPA有更高的选择性和回收率。和CE相结合,BPA的回收率为95.20%~105.40%,检出限为1.8 μg/L。随后,该课题组[36]又提出了一种更快捷的在线分子印迹SPE(on-line MIP-SPE)连接CE进行检测复杂样品(诸如自来水、河水、饮料和尿样)中的BPA含量。将MIP材料包裹于分离毛细管上,制备了一种柱内MIP-SPE浓缩器,在最优条件下,线性范围为(3~300)ng/mL,检出限为0.8 ng/mL。和报道的离线MIP-SPE-CE方法相比,在线方法能够实现在线的提取和去吸附。与其他检测器相比,MS能够得到更明确的结果。Zeng等[37]用CE-飞行时间质谱(CE-TOF/MS)检测鼠尿中极性和离子型的代谢物。过程为:离心除去样品中的蛋白质,用CE-TOF/MS分析尿样中的亲水性提取物。通过同轴鞘液界面[50%甲醇/水,(v/v)和0.1 μM苯丙烯磷腈]实现了CE-MS联用。尿样先通过熔融的石英毛细管在20 ℃下进行分离,然后用TOF-MS在阴离子和阳离子条件下分别进行分析测定。该研究认为,CE-MS分析方法的多元模式识别能够直接反映BPA造成的代谢紊乱。Mei等制备了一种BPA-分子印迹聚合物微球,将其用作BPA萃取的SPE吸附剂。将该方法与CE相结合,分别开发了离线[35]和在线[36]的方法,用于测定水样、土壤和尿样中的BPA含量。离线方法中,BPA的回收率为95.20%~105.40%,检出限为1.8 μg/L。在线方法在最优条件下,线性范围为(3~300) ng/mL,检出限为0.8 ng/mL。
2.4 免疫分析技术(immunoassay) 2.4.1 酶联免疫吸附测定(ELISA)ELISA是将高特异性抗原抗体相互作用和酶的有效催化相结合的一种免疫分析技术。ELISA具有高特异性、高敏感度和高通量等优势,近些年发展迅速。Lu等[38]提出了一种直接和间接竞争的ELISA,基于一系列多克隆抗体和不同的竞争半抗原。Lei等[39]提出并比较了直接和间接竞争性ELISA检测尿样中BPA的方法,结果显示直接竞争ELISA特异性和灵敏度均更高。直接和间接法的检出限分别为0.03和0.08 ng/mL。Maiolini等[40]用三种方法评价了塑料婴儿奶瓶中BPA的迁移,分别为测流免疫测定、间接竞争化学发光酶联免疫吸附测定(CL-ELISA)和LC-MS/MS。结果显示CL-ELISA的IC50和检出限分别是0.2和0.02 ng/mL。LC-MS/MS和测流免疫测定的检出限分别为5.8 ng/mL和0.1 μg/mL。和LC-MS/MS相比,CL-ELISA具有更敏感、更快捷和高通量筛选的优势。
2.4.2 免疫层析法(immunochromatographic assay)免疫层析试纸条是一种经典的快速检测手段,具有低成本、易操作、用户友好等特点。Mei等[41]将抗BPA抗体和金纳米颗粒(AuNPs)相连,制备了新型免疫层析试纸条,用于检测BPA。抗BPA抗体和金纳米颗粒作为识别探针,BPA-牛血清连接物和抗鼠抗体分别作为测试线和控制线。利用该方法可以快速灵敏地筛查水样中的BPA,通过肉眼和软件分析最低可测5 ng/mL和0.92 ng/mL,为快速筛查检测BPA提供了可行性。为提高检测的灵敏度,该课题组又制备了一步信号放大的超灵敏BPA检测免疫层析试纸条[42]。将抗牛血清白蛋白抗体标记物包裹于颗粒较大的金纳米颗粒上,通过免疫反应来增强信号。方法的敏感度提高了至少10倍,肉眼检测BPA的检出限为0.5 ng/mL。
2.5 基于传感器的分析方法根据不同的识别元件来分类,可大致分为化学传感器和生物传感器。目前研究发展了很多关于BPA的化学传感和生物传感方法[43]。
2.5.1 化学传感器化学传感器主要包括电化学传感器和化学发光传感器。其中,分子印迹聚合物传感器是发展最多的电化学传感器。分子印迹技术是模拟抗原-抗体反应机理,在聚合物材料中引入分子识别位点。是目前发展高选择性材料的方法之一。以MIPs为基础构建的传感器,对BPA有特异性,因此可作为BPA的识别因子。Xiong等[44]制备了一种以MIP为基础的光纤倏逝波传感器,用于检测BPA。BPA被印迹膜识别,被纤芯表面产生的倏逝波激发产生荧光,用荧光检测器进行检测,该方法检出限为1.7×10-9 g/mL。Zhu等[45]在磁性纳米颗粒基础上修饰了MIPs,合成了新型的MMIP传感器。该传感器的检出限为1.0×10-7 mol/L。Ensafi等[46]基于金纳米颗粒(AuNPs)和玻碳电极(GCE)制备了用于BPA检测的电极。在最佳条件下,该方法的BPA检测浓度范围为0.5 fmol/L到5 pmol/L,检出限为8.0×10-17 mol/L,应用于检测牛奶、奶粉和水样。
2.5.2 生物传感器为了提高电化学检测的选择性,很多生物分子,包括抗体[47]和核酸适配子都被用作识别因子。抗体生物识别分子具有很大的优势,然而抗体非常昂贵,难修饰也很难将其固定。抗体是将分析物分子注射入生物体内而产生的,不仅耗时费力,而且价格昂贵。因此,核酸适配子是抗体的优良替代品。核酸适配子是合成的单链寡核酸或肽分子,也被称为化学抗体。由于它们相似的识别属性但各不相同的化学性质,核酸适配体被认为是传统抗体的潜在替代物,并被用于检测一系列化合物[48]。自20世纪90年代起,适配子已经展示出它的优点,诸如合成简单,成本低,体积小,反应灵敏,特异性等[49]。这些年,各种核酸适配体不断涌现,基于BPA的适配子结合了荧光光谱、比色法、电化学和表面增强拉曼光谱(SERS)等多种技术[50]。Kuang等[51]制备了一种非对称等离子体手性适配体传感器用于BPA检测。抗BPA适配子及其互补序列分别用20 nm和10 nm的金纳米颗粒修饰。在无BPA条件下,互补序列和适配子通过DNA诱导杂交作用形成不对称等离子体纳米颗粒二聚体。而在BPA存在时,适配子更倾向于和BPA相结合,使二聚体分解成为分散的纳米颗粒。在最佳条件下,该方法的检出限为0.008 ng/mL。Chung等[52]制备了一种表面增强拉曼散射适配体传感器,能够超灵敏痕量分析检测水样中的BPA,该方法的检出限为10 fmol/L。Zhu等[53]用氧化石墨烯和荧光改性的适配子,制备了一种荧光共振能量转移传感器,用于一步检测BPA。抗BPA适配子用羧基荧光素、氧化石墨烯作为荧光淬灭剂。荧光信号的强度和BPA的浓度成正比。该方法的检出限为0.05 ng/mL, 在(0.1~10) ng/mL呈良好线性关系。Kashefi-Kheyrabadi等[54]通过在电化学传感器上插入特殊设计的微通道,组成几何激活表面相互作用芯片,是第一个用于检测BPA的微流控集成电化学传感器(MEA)。该方法线性动态范围较宽:(5×10-12~1×10-9) mol/L,检出限是2×10-13 mol/L。Lee等[49]以金纳米胶体颗粒和BPA特异性适配子制备了BPA比色传感器,当检测到BPA分子时,适配子与BPA发生电解质诱导聚合,纳米颗粒的表面等离子体共振位移使得产生肉眼可见的由红到蓝的颜色变化。肉眼可观测到的BPA最低浓度为1 pg/mL。
3 总结与展望BPA作为化工行业的中间体,在全球范围广泛分布,对环境的污染越来越严重,引发全世界范围关注。环境和生物样品中的BPA前处理和检测方法都已取得诸多进展。前处理方面,微萃取技术的发展促进了样品处理的绿色化和人性化。选择性吸附剂,如MIPs的发展,将分离和除杂过程进行融合,提高了特异性和分离效率。这些方法都还处于发展中,但都展现了不错的前景。总之,样品制备过程正朝着更简便有效、更环保、自动化的方向发展。检测方面,色谱和质谱仍是广泛应用的技术手段,考虑到质谱分析需要复杂的前处理过程,高选择性材料(如MIPs、MMIPs和抗体等)的研制和大量应用商业化,将会大幅提高检测效率。ELISA和免疫层析法是目前BPA检测最常用的免疫分析技术,主要的缺点是该分析技术只能进行半定量分析,且常常伴随假阳性。适配体传感器和MIP传感器在检测BPA方面展现了巨大的潜能。随着纳米技术的高速发展,不同的纳米材料的应用将是分析检测BPA的另一个重要趋势。
