植物寄生线虫是一类世界性分布的重要植物病原物,其寄主范围广,环境适应性强,几乎可以危害所有农作物。目前已报道的植物寄生线虫种类约4 000余种[1],每年在世界范围内造成的损失超过800亿美元[2]。其中危害严重的种类有固着性内寄生的根结线虫(Meloidogyne spp.)、孢囊线虫(Heterodera spp.)和球孢囊线虫(Globodera spp.);另外短体线虫(Pratylenchus spp.)、穿孔线虫(Radopholus similis)、起绒草茎线虫(Ditylenchus dipsaci)、松材线虫(Bursaphelenchus xylophilus)、肾形肾状线虫(Rotylenchulus reniformis)、标准剑线虫(Xiphinema index)、异常珍珠线虫(Nacobbus aberrans)和水稻干尖线虫(Aphelenchoides besseyi)也属于危害较严重的线虫[3]。我国地处温带和亚热带,气候条件良好,适宜于线虫的活动和繁殖。上述的重要植物寄生线虫除异常珍珠线虫外,其余在我国均有报道[4]。
1.2 植物寄生线虫效应蛋白的研究概况植物寄生线虫是专性活体营养寄生物,它们运用各种手段与寄主植物发生相互作用,如降解植物的细胞壁,抑制植物的防御反应,调控植物的信号通路等,从而有利于自己的侵染和寄生[5]。在侵染寄主植物的过程中,植物寄生线虫利用其中空的口针,穿刺植物的细胞壁,并通过口针将来自于3个食道腺细胞的分泌物注入到植物细胞中,诱导植物细胞发生形态和结构的改变,最终形成维持其整个生活史营养来源的取食位点[6]。了解这些分泌物的遗传和分子生物学特征,对研究植物寄生线虫的致病机理和寻找防治靶标具有重要意义。
在早期的研究中,将从这些分泌物中分离和鉴定到的基因称为寄生基因,而将这些基因编码的蛋白质称为分泌蛋白[7]。近年来在植物寄生线虫研究中,效应蛋白被研究者广泛接受并应用。效应蛋白最初是指所有能够改变寄主植物细胞结构和功能的病原物蛋白和害虫蛋白,以及小分子物质[8]。也有人将效应蛋白定义为能够抑制寄主植物的防御反应,而有利于病原物侵染的一类蛋白[9]。
自从1998年第一篇关于植物寄生线虫效应蛋白的研究报道以来[10],编码植物线虫效应蛋白的基因的克隆和功能分析已成为线虫学研究热点,且研究取得了很大进展。据报道,目前大约有超过100个植物寄生线虫的效应蛋白其编码基因被克隆和研究,有些蛋白的功能和与寄主植物的相互作用已经阐明[5],其中研究最多的是植物细胞壁降解酶一类的效应蛋白,其次还有抑制植物防御反应和促进线虫取食位点形成的效应蛋白。然而,绝大多数植物寄生线虫的效应蛋白功能还是未知的[11]。
2 植物寄生线虫效应蛋白功能分析的主要方法 2.1 RNA干扰RNA干扰(RNA interference,RNAi) 是指外源或内源的双链RNA(dsRNA)特异性地引起基因表达沉默的现象。这种现象是生物为保护自身基因组免受外源(如病毒)和内源性(如转座子元件)序列的侵袭,而特异性调节或干扰基因表达的一种自身防御免疫应答现象[12]。由于植物寄生线虫属于活体营养的专性寄生物,不能人工培养和创造突变体库,缺乏成熟的转基因方法,这些特点都使得常规的正向遗传学研究方法在植物寄生线虫中受到很大限制,RNAi作为一种反向遗传学技术,为研究植物寄生线虫效应蛋白功能提供了极大便利[13]。RNAi首先是在秀丽小杆线虫(Caenorhabditis elegans)中发现,随后在植物寄生线虫研究中得到了广泛应用[14]。对于植物寄生线虫,实现RNAi最主要的方法是浸泡,通过在浸泡的dsRNA溶液中加入章鱼胺、间苯二酚、血清素和卡巴胆碱等神经兴奋剂,刺激2龄幼虫取食溶液中的dsRNA,得到RNAi效果,从而观察靶标基因沉默后植物寄生线虫的表型变化,推测靶标基因在线虫侵染和寄生过程中的作用。在植物寄生线虫中,已经通过体外RNAi方法对多个基因进行了基因沉默,初步推测了这些基因的功能。这些基因主要来自于危害最严重的孢囊线虫、球孢囊线虫、根结线虫、松材线虫和香蕉穿孔线虫 [13, 15, 16, 17, 18, 19, 20, 21, 22, 23, 24, 25]。
虽然利用体外浸泡方法可以产生很好的RNAi效果,但是存在的缺点是通过浸泡获得的RNAi效果是短暂的,为了获得持久的RNAi效果,利用寄主植物传递dsRNA到寄生阶段的线虫体内成为研究线虫效应蛋白功能的重要手段,同时也成为研究抗线虫转基因的热点。植物介导的RNAi在根结线虫和孢囊线虫中已得到很好应用,且获得了良好的抗线虫植株。比如在含有寄生性基因16D10发夹式结构的转基因拟南芥(Arabidopsis thaliana)上接种根结线虫,根结的数量减少了63%~90% [26];利用植物介导的RNAi沉默甜菜孢囊线虫(H. schachtii)的靶标基因10A06等几个基因后,孢囊线虫对转基因拟南芥的侵染率显著下降[27]。通过转基因烟草(Nicotiana benthamiana)沉默爪哇根结线虫(M. javanica)的假定转录因子MjTis11后,检测到接种在转基因烟草上的线虫体内MjTis11显著下调,并且在转基因的烟草中检测到siRNA,这充分说明了在转基因的烟草中产生了RNAi的效果 [28]。
近年来利用病毒介导的植物体内RNAi也在植物寄生线虫效应蛋白功能研究中得到广泛应用。烟草脆裂病毒(tobacco rattle virus,TRV)因为在根内能够迁移而且不产生有害的症状而被广泛应用,在甜菜孢囊线虫、南方根结线虫(M. incognita)和爪哇根结线虫中都得到了良好的应用[29, 30, 31]。这种方法的优点在于可以快速研究效应蛋白在植物体内的RNAi效果,缺点在于不能产生稳定的、可遗传的RNAi转基因植株,在抗线虫基因工程中受到了一定限制。
2.2 在植物中表达线虫效应蛋白在植物中表达线虫效应蛋白通常称为过表达,这也是植物寄生线虫效应蛋白功能研究中常用的一种方法。通过在植物中瞬时表达或稳定表达相应蛋白,观察过表达后植物表型变化,以及植物对线虫侵染和寄生的影响,可以推测效应蛋白的功能。在植物中过表达线虫效应蛋白通常与植物介导的RNAi相结合,从而分析线虫效应蛋白在线虫寄生过程中的功能和作用。
目前通过过表达技术已对一些植物寄生线虫的效应蛋白开展了功能研究。例如,在拟南芥中稳定过表达甜菜孢囊线虫的纤维素结合蛋白后,能够促进拟南芥根系生长,转基因拟南芥根系与野生型相比,显著增长,并且提高了拟南芥对甜菜孢囊线虫的感病性,通过这些结果可以推测甜菜孢囊线虫的纤维素结合蛋白对线虫侵染起着非常重要的作用[32]。类似的研究还有甜菜孢囊线虫的10A06和19C07,爪哇根结线虫的MJ-NULG1a和MJ-FAR-1,南方根结线虫的8D05等效应蛋白[30, 33, 34, 35, 36]。
2.3 效应蛋白的时空表达模式 2.3.1 效应蛋白在线虫体内的组织定位植物寄生线虫在侵染寄主植物过程中分泌许多效应蛋白到植物细胞中去,发挥各种各样的作用,从而保证线虫的寄生和繁殖。植物寄生线虫的食道腺是效应蛋白产生的主要器官,另外还有头感器、皮层组织、尾感器等器官[37]。不同器官来源的效应蛋白在植物寄生线虫寄生过程中发挥的作用也各不相同,研究效应蛋白在线虫体内的组织定位,有助于对效应蛋白开展更深入的功能研究。一般通过原位杂交或免疫定位的方法来研究效应蛋白在线虫体内的表达定位,通过合成特异性探针或制备特异性抗体,与固定后的线虫进行杂交,显色后观察杂交信号的位置。在固着性内寄生植物线虫中,亚腹食道腺产生的效应蛋白主要在线虫侵染早期迁移阶段发挥作用,而背食道腺产生的效应蛋白主要对线虫取食位点形成和维持起作用[7]。细胞壁降解酶类的效应蛋白是植物寄生线虫中研究最多、最重要的一类效应蛋白,通过原位杂交或免疫定位方法研究表明,大多数细胞壁降解酶基因定位于线虫的亚腹食道腺,主要用于降解植物细胞壁,在线虫的入侵或迁移阶段发挥作用[37]。线虫头感器也是重要的分泌器官,它暴露于周围环境之中,可以直接将效应蛋白分泌到植物组织中[38]。目前定位于头感器的线虫效应蛋白发现较少,如MAP-1是从南方根结线虫无毒种群中分离出来的一个效应蛋白,定位于南方根结线虫头感器,推测其在线虫与植物互作的早期识别阶段起作用[39]。线虫尾感器与头感器结构相似,也可以分泌效应蛋白,从南方根结线虫侵染性2龄幼虫的分泌蛋白组中分离到一个CDC48类蛋白(CDC48-like protein),与秀丽小杆线虫的CDC48蛋白高度相似,定位于线虫的尾感器,可能通过调节植物细胞的分裂和增殖而有助于取食位点的形成[40]。定位于线虫皮层组织的效应蛋白也比较少,研究表明马铃薯金线虫(G. rostochiensis)的SXP-RAL2蛋白、水稻干尖线虫的Ab-FAR-1蛋白定位于线虫的皮层组织,这可能是协助线虫从寄主组织中吸收维生素A而维持自身的代谢,也可能参与抵抗植物的防卫反应而完成侵染[41, 42]。
2.3.2 效应蛋白在线虫不同发育阶段的表达编码植物寄生线虫效应蛋白的基因在线虫不同发育阶段发挥不同作用,其表达量也有所差异,根据特定基因在不同发育阶段的表达模式,可以推测其在线虫寄生过程中的作用,也是研究效应蛋白功能的基础。早期用半定量的方法来检测基因在不同虫态的表达量,而实时荧光定量PCR能够更加准确地反映基因在线虫不同发育阶段的表达量,其结果通常与效应蛋白在线虫体内的组织定位相结合来分析效应蛋白的功能。
在固着性内寄生线虫侵染性2龄幼虫阶段表达量较高的基因,其编码的效应蛋白主要在侵染的早期迁移阶段发挥作用,而在寄生阶段表达量高的基因,主要是对取食位点的形成和维持起调节作用。细胞壁降解酶类效应蛋白主要用于降解植物的细胞壁,在线虫侵染过程中发挥主要作用,编码这一类效应蛋白的基因在侵染性2龄幼虫阶段的表达量高度上调[18, 43, 44, 45]。南方根结线虫效应蛋白8D05、爪哇根结线虫效应蛋白MJ-NULG1a、甜菜孢囊线虫效应蛋白30C02和马铃薯金线虫效应蛋白GrUBCEP12都是在线虫寄生性2龄幼虫阶段表达量最高,因此认为线虫可能通过分泌这些效应蛋白,来改变植物细胞结构,促进取食位点形成,以保证寄生阶段的完成[30, 36, 46, 47]。
2.4 线虫效应蛋白在寄主细胞中的定位固着性内寄生的根结线虫和孢囊线虫侵染寄主后,通过分泌效应蛋白,调控植物细胞形态、功能和基因表达水平变化,从而诱导植物细胞形成高度分化和代谢活跃的取食位点,作为线虫完成生活史的唯一营养来源[5]。通过亚细胞定位和组织化学切片方法,可以观察效应蛋白在植物体内的作用位点,分析效应蛋白在调控植物细胞分化代谢过程中的作用。亚细胞定位的研究需要构建带有特异性效应蛋白与荧光蛋白(GFP)相融合的瞬时表达载体,通过基因枪法、农杆菌转染法或转化植物原生质体的方法,在植物细胞中瞬时表达后,根据荧光信号的位置,判断效应蛋白的分泌特性及其在植物细胞中的定位情况。研究表明,南方根结线虫效应蛋白MI-EFF1和7H08、爪哇根结线虫效应蛋白MJ-NULG1a和甜菜孢囊线虫效应蛋白19C07和10A07都定位于植物的细胞核,可能通过调控植物细胞的基因表达水平或细胞周期循环影响线虫的寄生和发育[30, 48, 49, 50, 51]。马铃薯金线虫效应蛋白GrUBCEP12定位于植物细胞质,可能通过分泌泛素蛋白来抑制基础防卫反应而促进合胞体形成 [46]。南方根结线虫效应蛋白MI-CRT则是定位于植物外质体,能够抑制植物防卫反应,在线虫与植物相互作用中有利于线虫寄生[52]。
组织化学切片是检测线虫效应蛋白在植物体内定位更加直接的方法。通过对包含有寄生线虫的植物组织进行固定和组织切片,与相应效应蛋白的抗体进行识别,通过显色后,观察免疫组织定位的信号。例如,爪哇根结线虫侵染番茄(Lycopersicum esculentum)5d、10d和18d时进行组织切片,用效应蛋白MJ-NULG1a的抗体进行检测,在番茄的细胞核内观察到稳定的分泌信号,因此确认MJ-NULG1a作用于植物细胞核[30]。采用同样的方法对定位植物细胞核的南方根结线虫效应蛋白MI-EFF1和甜菜孢囊线虫效应蛋白19C07做了进一步验证,结果与亚细胞定位结果一致[35, 49]。另外,对爪哇根结线虫的脂肪酸和维生素A(MJ-FAR-1)结合蛋白进行免疫定位研究发现,在寄生爪哇根结线虫的拟南芥根内,在线虫表皮及线虫与植物细胞的连接处观察到明显的MJ-FAR-1分泌信号,表明MJ-FAR-1可分泌到植物体内,在整个寄生阶段与植物发生相互作用,有利于线虫的寄生和繁殖[34]。
2.5 线虫效应蛋白的受体蛋白筛选与功能分析植物寄生线虫在侵染和寄生过程中,通过分泌效应蛋白与寄主蛋白进行相互作用,来保证自己的侵染和寄生。因此,可以通过寻找效应蛋白在寄主体内的受体蛋白,进而通过分析受体蛋白功能来推测线虫效应蛋白的功能。目前通常采用酵母双杂交方法来筛选受体蛋白。在植物寄生线虫中,一般通过构建线虫效应蛋白的诱饵载体和寄主植物的cDNA文库,把诱饵载体和cDNA文库转化到酵母细胞中,在合适条件下进行杂交,通过检测报告基因来筛选蛋白之间的相互作用[54]。当然,酵母双杂交有可能获得假阳性受体蛋白,因此通常需要双分子荧光互补技术(Bimolecular fluorescence complementation,BIFC)或免疫共沉淀(Co-immunoprecipitation,Co-IP)等方法进行验证。已经通过BICF方法或Co-IP方法验证了一些线虫效应蛋白与植物蛋白之间的相互作用[32, 33, 35, 48]。例如甜菜孢囊线虫纤维素结合蛋白与拟南芥果胶甲基酯酶存在相互作用[32];甜菜孢囊线虫效应蛋白10A06与拟南芥亚精胺合成酶2、10A07与拟南芥的IAA16转录因子、19C07与拟南芥生长素转运因子LAX3之间的相互作用也已经得到证明[33, 35, 48]。
在获得受体蛋白后,通过进一步对受体蛋白的研究,将有助于对线虫效应蛋白功能的分析。在拟南芥中过表达线虫效应蛋白30C02的互作蛋白AT4G16260,转基因拟南芥对甜菜孢囊线虫的感病性下降,而拟南芥的AT4G16260突变体对甜菜孢囊线虫的感病性明显提高[47]。而过表达10A06的互作蛋白SPDS2后,转基因拟南芥对甜菜孢囊线虫的感病性上升,而SPDS2的突变体对甜菜孢囊线虫的感病性却有所下降[33]。过表达WRKY转录因子,甜菜孢囊线虫在转基因拟南芥上形成的合胞体直径,比突变体拟南芥根内线虫形成的合胞体直径要小很多,侵染率也下降很多,因此推测,线虫可能通过干扰植物体内毒素转运来创造自身的寄生条件[56]。此外,在大豆根内过表达拟南芥的两个α-内切-β-1,4-葡聚糖酶基因AtCel6和Gmcel7后,线虫在转基因大豆根内的发育均受到限制,这也说明线虫可能通过抑制植物体内纤维素合成来促进自身发育[57]。
酵母双杂交系统在根结线虫中应用相对较少,已有的研究表明,南方根结线虫效应蛋白16D10与拟南芥两个转录因子AtSCL6 和 AtSCL21相互作用,这也是首次研究证实植物寄生线虫的效应蛋白能够特异地作用于植物的调控蛋白,可能作为信号分子调控拟南芥根的分化发育[55]。南方根结线虫的另一个效应蛋白8D05则与番茄液泡膜上的水通道蛋白相互作用,通过调节巨型细胞内物质和水分运输而促进线虫的寄生[36]。这些研究结果对于进一步分析效应蛋白的功能至关重要。
2.6 效应蛋白与植物的防卫反应植物对病原物的防卫反应分为PTI(PAMPs-triggered immunity)和ETI(Effector-triggered immunity)。PTI由病原物相关分子模式(pathogen-associated molecular patterns,PAMPs)引起。PTI反应主要引起植物体内活性氧产生、细胞内胼胝质沉积和防卫相关基因表达量变化等。ETI是效应蛋白被植物抗病蛋白识别后引起的免疫反应,ETI反应导致在侵染点附近产生过敏性反应(Hypersensitive reaction,HR)[58]。爪哇根结线虫和大豆孢囊线虫的分支酸变位酶是关于植物寄生线虫效应蛋白抑制植物防卫反应最早的报道,推测线虫可能通过产生分支酸变位酶,与植物竞争分支酸,导致水杨酸和植保素合成受到抑制来调控寄主的防卫反应[59, 60]。对南方根结线虫侵染后的拟南芥根组织进行转录组研究表明,根结内防卫基因的表达量发生显著变化,这说明线虫取食位点的形成与抑制植物防卫反应有关,而这些上调或下调的基因都属于植物次级代谢或受压情况下表达的基因,这属于典型植物防御病原物的机制[61, 62, 63, 64]。另外甜菜胞囊线虫效应蛋白Hs10A06可能影响植物的水杨酸途径,从而抑制植物的基础抗病性[33],大豆孢囊线虫效应蛋白Hg30C02作用于一个病程相关蛋白而参与抑制植物抗病性[47]。
以上研究都只是推测效应蛋白在植物防卫反应中的作用,没有直接的证据。近几年,植物病原物效应蛋白抑制植物防卫反应的方法开始在植物寄生线虫效应蛋白功能研究中得到应用。在研究植物寄生线虫效应蛋白与PTI反应的关系时,一般以细菌鞭毛蛋白N端的22个氨基酸(flg22)或细菌中高度保守的18个氨基酸肽段(elf18)作为PAMPs。在烟草叶片中瞬时表达线虫的效应蛋白后,用flg22或elf18等PAMPs处理瞬时表达后的叶片,用化学发光仪检测叶片产生的活性氧ROS,用实时荧光定量PCR检测防卫基因表达量的变化,通过苯胺兰染色观察细胞壁中胼胝质的沉积等。如:近期对南方根结线虫钙网蛋白(Mi-CRT)进行研究时发现,在拟南芥中瞬时表达Mi-CRT后,用elf18处理瞬时表达后的拟南芥,qRT-PCR检测显示,瞬时表达Mi-CRT的拟南芥其体内防卫相关基因表达量与对照相比显著下降,而且叶片细胞壁胼胝质沉积量显著减少,这说明瞬时表达Mi-CRT的拟南芥植株抑制了由elf18诱导的PTI反应,导致病程相关基因表达量下降,胼胝质减少[52]。这是第一个被证明具有抑制PTI反应功能的植物寄生线虫效应蛋白。另外,在烟草中瞬时表达马铃薯金线虫泛素蛋白的GrCEP12后,能够抑制由flg22诱导的PTI反应,主要表现在抑制了烟草体内ROS产生和防卫基因表达[65],说明GrCEP12与寄主防卫反应有关。
ETI是由效应蛋白激发的植物防卫反应。能够被植物体内抗性蛋白识别的效应蛋白称为无毒蛋白,无毒蛋白能够激发由植物抗性蛋白诱导的过敏性坏死反应等症状[58]。在植物寄生线虫中,关于无毒蛋白的研究相对较少,目前的研究推测南方根结线虫的MAP1和爪哇根结线虫的Cg1效应蛋白可能是潜在的对抗番茄抗性基因Mi-1的无毒蛋白,但是还需要通过确凿的实验数据来证实[16, 39]。马铃薯白线虫(G. pallida)的Gp-RBP-1是仅有的一个被证实的植物寄生线虫无毒蛋白[66]。近年来,利用无毒蛋白Gp-RBP-1与抗性蛋白Gpa2在植物体内瞬时表达后产生的过敏性坏死反应来研究线虫效应蛋白的功能得到较多应用。最近报道的马铃薯金线虫效应蛋白Gr-SPRYSEC-19和Gr-UBCEP12都能够抑制由Gp-RBP-1/Gpa2或CP/Rx1诱导产生的细胞程序性死亡,这说明Gr-SPRYSEC-19和Gr-UBCEP12可能具有抑制植物ETI的功能[46, 67]。
3 总结与展望植物寄生线虫属于专性活体寄生物,这给植物寄生线虫效应蛋白的分离和功能研究带来了很大困难。在真菌和细菌中常用的一些分子生物学技术在植物寄生线虫中难以实现,因此,植物寄生线虫效应蛋白的功能研究相对比较缓慢。
虽然已经获得大量的植物寄生线虫效应蛋白,但是只有少数的效应蛋白功能得到研究和分析。南方根结线虫、北方根结线虫和松材线虫等植物寄生线虫基因组测序的完成和转录组测序在植物寄生线虫效应蛋白研究中的广泛应用[68, 69, 70, 71, 72],为分离植物寄生线虫效应蛋白提供了很好的平台,目前急需要对大量未知功能的植物寄生线虫效应蛋白进行功能研究。
在现有的这些植物寄生线虫效应蛋白功能研究技术水平上,结合生物信息学分析,以及借鉴研究其他植物病原物效应蛋白功能的方法,将会有更多植物寄生线虫效应蛋白的功能得到研究和分析,效应蛋白在植物中的作用靶标和参与调控的植物信号通路将成为植物寄生线虫效应蛋白研究中的重点和热点,这将为人们更加深入地了解植物寄生线虫与植物相互作用的分子机理奠定基础,为寻找可持续的植物寄生线虫防控策略提供理论依据。
