危害植物的有害真核生物主要有真菌、卵菌、草食昆虫、寄生线虫和杂草等,其中大多数病虫害的发生和流行会造成作物产量的巨大损失和品质劣变,一些毁灭性病害的发生还可能危及世界农业生产和粮食安全[1, 2, 3]。目前,农业生产中防制病虫害的措施主要有轮作倒茬、化学控制和培育抗病品种三种途径。合理的轮作制度及栽培措施的改进尽管可以在一定程度上减轻病虫危害[4, 5, 6, 7],但由于受种植业比较效益、种植习惯等因素的影响,这一措施在农业生产中无法得到有效利用。使用杀菌剂、杀虫剂等农药尽管可以较好控制病虫害的发生,但化学农药的大面积长时间使用,不仅会对人类健康造成危害,而且存在巨大的生态风险,危及我们赖以生存的环境,因此化学农药的使用不得不受到限制[8]。培育抗性品种是公认的控制病虫害最经济、最有效的途径,但传统抗性育种手段耗时耗力,并且天然抗性资源十分有限,限制了抗性品种选育的进程[9, 10]。转基因育种可以突破物种间的界限,利用其他物种的抗性资源增强作物抗性,可以一定程度拓宽抗性基因来源,加速育种进程。但由于许多病原物和害虫具有极强的进化潜力,能够迅速克服新的抗性基因,不但使采用传统抗性育种手段选育的抗性品种在短期内被克服,连转基因的Bt毒性蛋白也在投放生产后不久逐渐失去效用[11, 12, 13, 14, 15]。所以,作物抗病虫害育种中抗性资源不足、新品种育成速度缓慢等问题日渐严峻。故探索新的作物病虫害持久抗性育种策略势在必行。
RNAi自发现以后,作为一种强大的反向遗传学手段广泛应用于功能基因组研究[16],同时在植物抗病毒育种中也迅速得到成功应用,并培育了许多抗病毒作物[17, 18, 19, 20, 21, 22, 23, 24]。2001年,Escobar等[25]又成功应用RNAi技术增强了植物对细菌的抗性。随着对RNAi机理认识的不断加深,植物介导的RNAi也逐渐应用于增强植物对寄生线虫、草食昆虫、真菌、卵菌等真核有害生物抗性的研究。这些研究表明植物介导的RNAi可以沉默入侵真核生物中与寄主产生的dsRNA同源的基因,赋予寄主植物对入侵病、虫、草报复性精准打击的能力。RNAi作为一种新的植物持久抗病、抗虫育种策略的优势逐渐显现。
1 RNAi机制RNAi是一种真核生物中普遍存在、进化上高度保守的同源依赖型基因沉默现象,其基本原理是:细胞中长的dsRNA或前体pre-miRNA能激活细胞中RNase Ⅲ家族Dicer(或DCL)的活性,被其切割成21~25bp的小干扰RNA (siRNA)或miRNA(本文通称为小RNA,即sRNA,是引起RNAi干扰最主要的两种小RNA)[26, 27];sRNA与多蛋白复合体AGO-Piwi结合形成RNA诱导的沉默复合物(RNA-induced silencing complex,RISC)[28];RISC以sRNA的反义链为向导,识别同源基因,然后通过使RNA聚合酶Ⅱ释放、组蛋白乙酰化或DNA甲基化等途径引起相应基因转录水平基因沉默,通过mRNA剪切或阻止翻译引起转录后水平的基因沉默[29, 30, 31, 32, 33]。另外,sRNA还可以作为引物,在RDRP作用下,以互补mRNA为模板合成新的dsRNA,新合成的dsRNA被Dicer切割后形成次级siRNA,实现RNAi信号放大[34]。RNAi作为一种古老的基因沉默机制,是真核生物抵御病毒感染及抑制转座子活性、保持基因组稳定的重要途径,也在调控生物自身基因表达、控制生长发育进程中发挥着重要作用。最近的研究还发现,RNAi也参与了寄主植物与真核病原菌的互作,病原菌能够通过向寄主中分泌sRNA,劫持寄主的AGO1蛋白,选择性沉默寄主天然免疫相关基因表达而增强致病性[35, 36];而寄主植物也可以通过RNAi途径调控自身抗性基因的表达,增强抗病性[37],并且还有推测认为植物中编码的sRNA一部分的靶标基因可能就是害虫基因[38]。
RNAi参与植物与入侵真核生物的互作的前提是RNAi信号可以在两者之间传递。研究表明,作为RNAi信号的sRNA或dsRNA不但能够在同一生物的细胞间传递或通过输导组织进行远距离传输,实现RNAi的系统发生,也可以在密切接触的同一物种的不同个体或不同物种间转移,实现RNAi信号跨越物种的转移[39, 40, 41, 42, 43]。RNAi信号在同一生物中通过细胞间传递或通过输导组织在同一生物体内远距离运输导致生物体特定基因的沉默称为系统性RNAi;RNAi信号在密切接触不同生物个体间传递或生物体通过摄取环境dsRNA或siRNA引起的基因沉默称为环境性RNAi[44]。环境性RNAi是植物介导的RNAi增强其对有害真核生物抗性的理论基础,即植物中形成的以有害真核生物特定基因为靶标的RNAi信号被入侵的真核生物通过取食、吸器吸收等途径摄取后,触发入侵生物RNAi机制,引起相应基因沉默,从而赋予植物报复入侵真核生物、给与其精确打击的能力。
2 RNAi在抗植物寄生线虫研究中的应用植物寄生线虫能够侵染绝大多数栽培物种类,造成粮食、纤维和园艺作物产量及品质大幅度下降,据估计全球每年因线虫引起的作物产量损失超过1 250亿美元,其中根结线虫、孢囊线虫等固着性内寄生线虫危害最为严重[45]。这些寄生线虫在进化过程中形成了独特的侵染机制,入侵植物根系后向维管组织迁移,并选择性地向寄主根部分泌寄生蛋白,使其基因表达模式向着有利于线虫寄生的方向发生显著改变,引起细胞生理和表型变化,形成巨细胞(根结线虫)或合胞体(孢囊线虫),为线虫持续提供生长和繁殖所需的营养,造成作物根系受损,为其它根部病害发生创造条件。大多数作物对线虫抗性很差,主要采用化学方法防治。
Fire等[46]研究发现与反义RNA相比,dsRNA能够更有效地引起秀丽线虫同源基因的沉默,进一步的研究发现通过饲喂、注射和浸泡都可以将外源RNAi信号传递到线虫中[47]。又有研究证明,发生转录后基因沉默的植物的总RNA提取物注射到线虫后会引起线虫相应基因发生沉默。这些工作的开展为采用植物介导的RNAi减轻线虫危害提供了依据[48]。Gheysen等[49]的工作证明了在寄主植物内表达以根结线虫寄生基因为靶标的dsRNA可以引起寄主对线虫感染的抗性。随后,通过培育能够表达线虫特定基因dsRNA的植物以增强对线虫抗性的研究逐渐开展起来。Yadav等[50]的研究表明,表达根结线虫剪接因子(splicing factor)和整合酶intergrase(一个染色体重构蛋白a chromatin remodelling protein)特异dsRNA的转基因植物能够成功下调线虫相应基因表达,表现出对根结线虫的强烈抗性。Huang等[51]采用植物介导的RNAi成功下调了4个根结线虫近缘种中一个刺激植物根系生长的线虫分泌多肽[secretory peptide (16D10)]编码基因,增强了植物对线虫的抗性,抗性水平介于63% 到 90% 之间。转基因烟草表达以根结线虫锌指蛋白样转录因子编码基因为靶标的dsRNA能够有效控制线虫的危害。进一步研究发现,植物表达根结线虫转录因子基因的dsRNA能够下调寄生线虫该基因的表达,但转录因子功能的丧失对线虫并没有产生不良影响[52]。在抗根结线虫取得进展的同时,对RNAi信号吸收能力较差的孢囊线虫抗性研究也取得了良好进展。Steeves等[53]培育的表达孢囊线虫主要精子蛋白基因dsRNA的大豆不但能够使寄生线虫繁殖能力大大降低,每克根组织中线虫的产卵量下降68%,并且还会影响到线虫后代的繁殖能力,每克根组织产卵量下降75%。Sindhu等[54]的实验证明表达4个线虫寄生基因的拟南芥可以使发育的雌虫的数量减少,在不同RNAi株系中雌虫减少量在23%~64%之间。不但寄主植物稳定转化表达的RNAi信号可以引起线虫基因沉默,病毒介导的基因沉默也可以通过寄主植物传递给寄生线虫。Dubreuil等[55]以对根结线虫胚胎发育和幼虫运动能力有重要作用的肌钙蛋白C基因Mi-tnc和参与线虫和寄主植物互作的寄生基因钙网蛋白的致病基因Mi-crt 为靶标,分别构建了烟草脆裂病毒(TRV)沉默载体TRV∷TNC和TRV∷CRT,农杆菌渗透感染烟草后接种根结线虫,发现根结线虫卵及后代幼虫中相应基因的转录本大幅度减少,并且接种了TRV∷TNC的烟草会使寄生线虫卵的孵化率降低了59%,而接种TRV∷CR虽然对已经成功寄生的线虫表型没有影响,但会影响后代的侵染寄主的能力,使线虫瘿数量大幅度减少。siRNA通过喂食后可进入感染幼虫的内部组织,触发沉默其寄生基因Mi-CRT(根结线虫食管腺的一个钙网蛋白基因),在幼虫时期Mi-CRT基因沉默影响线虫的毒力[56]。Huang等[57]克隆了线粒体ATP合酶b亚单位基因MiASB,使烟草脆裂病毒(TRV)介导的病毒诱导的基因沉默在南方根结线虫中表达,然后在番茄幼苗上接种线虫,60天后,经MiASB沉默处理的幼苗跟对照相比,线虫瘿减少了64.3%。当线虫吸收能引起系统RNAi反应的dsRNA或siRNA后,很大范围细胞类型的基因表达被沉默。现在线虫已经成为RNA干扰基因沉默的模式生物,Lilley等[58]总结了植物中以寄生线虫基因为靶标的dsRNA的应用。
3 RNAi在抗草食昆虫研究中的应用草食昆虫采食植物地上组织部分,减少了植物光合组织的面积,导致植物体内光合产物的减少,影响植株生长与繁育,甚至导致植物死亡,对农林牧业造成严重的经济损失。就植物而言,同一种植物可能受到多种类型昆虫的取食[59];有些草食昆虫的专食性特别强,只取食某种植物,这两种情况都会给植物带来非常大的危害。有研究报道温带森林的辽东栎叶片的被食频率竟高达90%[59]。虽然昆虫对植物的取食压力使得植物进化出多种防御机制,但是反过来昆虫为避免被饿死或毒死又进化出应对植物防御的机制[60],这样就使得植物防虫害工作进展更加困难。
近期采用RNAi技术控制作物虫害的研究进一步拓展了RNAi在作物改良中的应用范围。线虫基因特异性抑制是可以通过喂养dsRNA实现的,最初认为把这种方法应用到昆虫中是不可行的,但是研究发现应用RNAi技术,得到耐食草昆虫的植物才刚刚得以实现,这为防治害虫,培育新一代的昆虫抗性农作物开辟了道路[61]。昆虫吸收dsRNA至少有两个途径:基于线虫的SID-1蛋白质的跨膜通道介导的摄取机制和“替代”的内吞作用介导的吸收机制[39]。Sivakumar等[62]利用RNAi技术细胞系实验,将Sf21昆虫细胞经过修饰后表达HaAPN1(棉铃幼上皮氨肽酶N),然后经过HaAPN1dsRNA处理48小时后,HaAPN1表达的mRNA及其蛋白质较对照分别降低70%,导致HaAPN1表达细胞对苏云金芽孢杆菌的Cry1Ac杀虫蛋白的敏感性降低。与此同期,Mao等[38]培育了能表达棉铃虫cyp6ae14基因(该基因编码一种能够代谢棉酚的蛋白,赋予棉铃虫对棉酚的抗性)dsRNA的转基因烟草和拟南芥,棉铃虫取食转基因植物后肠中cyp6ae14基因转录本大幅度降低,使棉铃虫对棉酚的耐受力减弱,导致其营养不良,生长缓慢,甚至死亡。随后Mao等[63]培育了能表达棉铃虫cyp6ae14基因dsRNA的转基因棉花,在幼虫喂食转基因植物4小时后,CYP6AE14表达水平降低,CYP6AE14蛋白水平下降,这些结果表明dsCYP6AE14转基因棉花对棉铃虫抗性增强。棉酚诱导的细胞色素P450中至少有这5种(CYP321A1,CYP9A12,CYP9A14,CYP6AE11和CYP6B7)促使棉铃虫耐受溴氰菊酯,幼虫通过取食CYP9A14基因dsRNA的转基因棉花和拟南芥后,表现出对菊酯杀虫剂更加敏感[64]。Baum等[65]培育了表达与美国西部玉米根虫(western corn rootworm,WCR)液泡ATPase编码基因同源的dsRNA的转基因玉米,增强了玉米对WCR的抗性,减轻了WCR对玉米的危害。Wang等[66]利用转录组测序(RNA-seq)和数字表达谱技术(DGE-tag)相结合,获得亚洲玉米螟卵、幼虫、蛹和成虫四个发育时期相关基因的表达时期、表达量、代谢通路及基因功能信息,针对其中10个幼虫期特异表达的基因合成 dsRNA,采用直接喷洒的方法对亚洲玉米螟幼虫进行处理,其中有9个基因的5天校正死亡率在 73%~100%之间,利用Real-time PCR 的方法检测基因表达变化情况,结果显示dsRNA处理后靶标基因的表达均有明显的下调。通过合成荧光dsRNA进行亚洲玉米螟幼虫的点滴试验,证明了dsRNA能够穿透昆虫体壁进入体腔从而实现基因干扰效果。RNAi技术已经在其他鞘翅目叶甲虫、小菜蛾、斜纹夜蛾等十几种草食昆虫多个基因中得到成功应用[39]。最近的研究表明,如果昆虫的性信息素受体通过RNAi被沉默,可以阻塞交配或宿主植物寻求行为的通信系统,就可以有效地控制害虫种群,而不必直接杀死害虫,这是RNAi应用的另一前景[67]。以上说明,通过植物介导的RNAi沉默昆虫体内的特定基因表达,能够控制虫害的发生。并且与杀虫剂抗性有关的特定基因沉默能增加昆虫对杀虫剂的特异敏感性,这将有效减少使用化学农药和提高化学农药效率[68]。
4 RNAi在抗真菌研究中的应用对植物来说,大多数病害都是由真菌引起的,由真菌引起的粮食作物病害已成全球性粮食安全问题,如果能控制这种病害在五大粮食作物中的传播,全球每年可多养活6亿人。随着全球化时代人员和各种物品大范围流通,原来仅限于某地的真菌感染也更容易扩散,其趋势有加重迹象。现在水稻、小麦、玉米、马铃薯和黄豆这五大粮食作物,都出现了由真菌引起的病害,如稻瘟病、大豆锈病、小麦秆锈病、玉米丝黑穗病和土豆晚疫病[69]。
RNA干扰技术作为真菌分子生物学研究或遗传改造的工具具有特殊的意义,因为真菌中丝状真菌具有多核和发生非同源重组频率高的特点,难以用基因敲除手段进行改造。最近几年,以绿色荧光蛋白基因和红色荧光蛋白基因为报告基因,在植物病原真菌M.oryzae、Venturia inaequalis、Colletotrichum lagenarium及模式丝状真菌 N.crassa 中建立了RNA干扰方法,并利用RNAi研究这些真菌某些重要基因的功能[70]。Liu等[71]于2002年利用 hpRNA表达载体诱导担子菌类病原真菌Cryptococcus neoformans的CAP59和ADE2基因的沉默,这是RNAi技术在抗真菌研究中的首次运用。随后,在稻瘟病菌中使用RNA沉默系统分析增强型绿色荧光蛋白(eGFP)基因,在沉默转化后,发现绿色荧光蛋白mRNA的积累急剧减少[72]。Nicolás等[73]的研究结果表明,这两种白化和野生型菌株具有相同水平的未剪接carB的mRNA前体,而沉默菌株成熟carB的mRNA的量至少比野生型少6倍,卷枝毛霉基因沉默发生在转录后水平,影响成熟mRNA积累,但未影响转录的速率。Tinoco等[74]的研究也表明,将转gus基因的拟轮生镰刀菌接种到表达gus-dsRNA的转基因烟草后,拟轮生镰刀菌的转基因gus表达被沉默。Sun等[75]的研究数据表明,RNAi介导的acuD基因的沉默能减轻马尔尼菲青霉的毒性从而在慢性期能减轻感染马尔尼菲青霉裸鼠的真菌负担。Trippe等[76]研究表明双链RNA介导的CYP52L1基因的转录后沉默,很大程度上降低了Graphium sp. (ATCC 58400)这种真菌氧化烷烃和醚类的能力。Salame 等[77]综述了RNAi技术已在40多种真菌中得到成功应用。Mascia等[78]表明,RNAi可以不用植物中间介导,通过病毒诱导的基因沉默在植物病源真菌炭疽病acutatum(菌株C71)表达,但是基于植物病毒,用烟草的重组病毒载体直接感染烟草花叶病毒(TMV),用类似方法在携带绿色荧光蛋白(GFP)异位表达的重组烟草花叶病毒载体中观察到相应基因沉默的现象。
卵菌(Oomycetes)属于色菌界(Chromista),由于其形态特征和生活习性与真正的真菌(true fungi)相似,通常被称为真菌。许多卵菌是植物病原菌,能引起多种农作物、花卉等发生灾难性病害,如大豆疫霉菌(Phytophthora sojae) 引起的大豆疫霉根腐病、致病疫霉(Phytophthora infestans) 引起的马铃薯晚疫病和橡树疫霉(P. ramorum) 引起的橡树猝死病[79]。
van West等[80]将致病疫霉分泌型激发因子编码基因inf1的编码区反向和/或正向导入致病疫霉,导致转基因致病疫霉inf1基因的沉默,首次证明了致病疫霉中RNAi机制的存在;进一步对转基因致病疫霉与野生型致病疫霉通过细胞质体融合获得的异核体的研究发现,inf1不但在异核体中保持沉默,在异核体非转基因的后代中也保持沉默,说明致病疫霉中沉默信号能在不同细胞核间传递。Whisson等[81] 建立了基于原生质体培养的外源dsRNA介导的致病疫霉瞬时基因沉默技术体系,采用人工合成的长度为150~300bp的dsRNA转染致病疫霉,激发了致病疫霉RNAi机制,分别引起致病疫霉转基因gfp、内源基因inf1及cdc14的特异性沉默。Judelson等[82]将表达与致病疫霉nifc1基因同源的发夹型RNA的载体导入致病疫霉,引起nifc家族基因(nifc1,nifc2和nifc3)的沉默,导致转基因致病疫霉游动孢子的萌发率大幅度降低。随后,Grenville-Briggs等[83]人工合成分别与致病疫霉ces1,ces2,ces3和 ces4同源的4种dsRNA,转染致病疫霉原生质体后抑制了4个ces基因的表达,导致转染后致病疫霉细胞壁纤维素含量不到野生型的50%。Colditz等[84]利用RNAi技术敲除苜蓿PR10-1基因[苜蓿感染豌豆根腐病菌丝后感应类10病原相关(PR)蛋白基因],蛋白质组学分析MtPr10-1和另外5个PR-10型标注为脱落酸反应蛋白(ABR17s)基因也被有效沉默,跟对照相比,接种病原菌的苜蓿根部病菌的定植明显减少了,在转MtPr10-1i基因的根部也抑制了病原感染,凝胶分析也证实了苜蓿增强了豌豆根腐病耐受性。水霉寄生的酪氨酸酶基因SpTyr是水霉菌生物合成途径不可或缺的基因,原生质体体外合成SpTyr的dsRNA介导了基因沉默,酪氨酸酶活性下降,黑色素生成量减少[85]。Stamler等[86]将体外合成cdc14基因的dsRNA使用电穿孔法导入到辣椒疫霉,游动孢子的生产量显著下降。当植物生产dsRNA时辣椒疫霉侵染叶片的能力被抑制,游动孢子的生产量也减少约46%。该技术在疫霉研究中的成功应用及越来越多疫霉基因功能的鉴定使得RNAi用于控制卵菌的发生和流行,增强植物抗性这方面的应用越来越实用。总之,RNAi在植物真菌相互作用中以真菌特异性为靶标设计抗真菌剂,可以适用于广泛的抗真菌研究[87]。
任琴等在实验室构建了以致病疫霉单拷贝的翻译延伸因子tef1为靶标的RNAi载体、以多拷贝的gpi基因为靶标的RNAi载体、同时与晚疫病菌4个ces基因均同源的融合基因C1234为靶标的RNAi载体和以几丁质酶基因为靶标的RNAi载体,采用农杆菌介导法转入晚疫病易感马铃薯品种大西洋,进行转基因植株晚疫病抗性的分析鉴定。其中,采用重叠延伸PCR技术克隆同时与晚疫病菌4个ces基因均同源的融合基因C1234,构建了内含子连接的C1234反向重复序列植物表达载体,采用农杆菌介导法转化晚疫病易感马铃薯品种大西洋,经PCR和Southern杂交检测,获得了129个转基因株系。离体叶片接种病原菌后,有97个转基因株系发病速度明显慢于野生型,接种6 d后病斑大小和霉层厚度均明显小于对照,并且叶片感病部位没有出现失绿斑,而野生型产生了明显的失绿斑。实时定量RT-PCR分析发现,发病延缓的叶片上致病疫霉4个纤维素合成酶基因的表达水平明显低于野生型。研究表明转基因植株中产生的以晚疫病菌ces基因为靶标的dsRNA能够沉默致病疫霉相应基因表达,延缓发病进程。
5 RNAi在抗寄生杂草研究中的应用植物界高等寄生植物约有3000 ~ 5000种,可导致主要粮食作物和工业农作物减产30%~80%[88]。其中寄生杂草是影响农作物生长和导致农作物产量和质量下降的高等寄生植物之一;其主要特性为不能独立地进行光合作用制造养分,必须寄生在别的植物上靠特殊的吸器吸取寄主的养分而生活,且种类繁多,分布广泛,寄主范围广,从寄主吸取水分、矿物质和光合产物的同时降低寄主的生长和竞争能力,对寄主的破坏性极大。作物被寄生杂草寄生后主要表现为生长缓慢,还会出现不同程度的生物量降低甚至死亡[89]。同时粮食和果实品质显著降低,例如大麻列当(Phelipanche ramose) 侵染番茄后,番茄鲜质量、干质量、硬度、还原糖、可溶性固体物含量和抗坏血酸含量等降低,果色变差,严重影响果实品质,降低其市场价值[90]。
寄生杂草中经济危害最大的当属broomrapes(列当属,列当科),witchweeds(独脚金属,玄参)和菟丝子(菟丝子属)[91],所以对其研究也最多。有研究指出寄主抗寄生植物的机理与其抗病原菌机理相似[92],所以使用RNAi技术亦可以实现寄生杂草特定基因的沉默。Aly等[93]利用伴胞中表达绿色荧光蛋白(GFP) 的转基因番茄探索寄主和寄生杂草之间的蛋白质运动,实验发现转基因番茄被分枝列当寄生后,大量的 GFP从寄主植物的韧皮部转移到寄生杂草的韧皮部,最终积累于列当的结节与芽。列当科的半寄生植物Triphysaria versicolor能够从寄主植物莴苣中摄取RNAi信号分子,导致自身转基因gus的沉默,并且能将沉默信号传递给其寄生的另外一株莴苣[94]。甘露6-磷酸还原酶(M6PR)是甘露醇生物合成的关键酶,Aly等[95]将寄生杂草分枝列当的6-磷酸甘露糖还原酶(M6PR) 基因作为靶基因得到转基因番茄,转基因番茄产生同源dsRNA将列当科寄生杂草的M6PR基因沉默,定量 RT-PCR 分析显示转基因番茄植株上生长的埃及列当结节和地下芽的内源M6PR mRNA 的量减少60%~80%,继而甘露醇水平显著降低,番茄植株上坏死结节比例显著提高。de Framond等[96]以5个独脚金基因为靶标构建发夹型结构dsRNA载体转化玉米,观察发现寄生转基因玉米的独脚金生长速度表现出一定的差异,与对照相比具有不可再生力。Bandaranayake等[97]将Triphysaria versicolor的胞质乙酰-CoA羧化酶(ACC酶)基因发夹结构RNAi载体导入苜蓿,在所有样本中均发现当列当科杂草寄生转基因寄主时其根的生存能力至少减少80%,RT-PCR表明Triphysaria中ACC酶转录水平显著下降。这项工作表明,ACC酶是抑制寄生植物反式特异性基因沉默的有效靶点。最近的研究表明,烟草脆裂病毒(TRV1和TRV2)诱导的S. hermonthica八氢番茄红素去饱和酶(PDS)基因的RNAi载体能有效沉默独脚金的同源基因,导致PDS基因在独脚金中的表达量下调,漂白其叶片[98]。虽然寄生杂草几十年都是棘手的问题,但是目前在基因组学上的进步将给这类研究带来一场革命,加快其前进的步伐[99]。通过RNAi技术降解寄生杂草靶mRNA并导致致命性伤害或抑制寄生植物的发展,从而控制杂草疯长防止其危害农作物,对稳定农作物产量、促进农业生产和保障世界粮食安全意义重大。
6 展 望植物地上部与地下部病原物如细菌、真菌、病毒和线虫等对植物叶片与根系的直接取食、损伤等不仅直接或间接影响植物的生长发育[100],更对粮食产量及果蔬质量造成严重影响。有害真核生物已严重影响了世界各地的农业生产力,每年造成巨大的农作物产量损失。植物介导的RNAi在农作物病虫害防治方面具有广泛的应用潜力。同时,也可以通过RNAi控制植物病原菌毒素的生长从而提高食品安全性。RNAi是一种高频、高效、高度特异的基因沉默技术,但是其干扰效率却有待提高,主要原因可能是当植物病原体渗入植物细胞内吸取营养成分时能有效避开植物自身的防御监测机制,从而不能让植物立即作出防御反应激发入侵有害真核生物的RNAi机制从而影响了干扰效率。植物介导的有害真核生物RNAi是通过在植物中形成dsRNA或siRNA等RNAi信号分子,经寄生、取食等途径传递到有害真核生物中,病原真菌入侵后如何使植物快速有效地发现并积极做出反应激发RNAi机制则需要更加深入的研究。
