动物来源放线菌的次级代谢产物及其生物活性研究进展

摘要: 放线菌是抗生素的主要来源之一,在人类对抗由致病菌产生的感染性疾病的过程中发挥了不可替代的防治作用。近年来,由于耐药菌株的不断涌现,促使人们加快寻找新的抗生素的步伐。寄生于动物体内的共生放线菌,是目前药用微生物资源的一个重要来源,也是当前微生物药物研究领域的一个热点。这些微生物在与动物互生共存的演化过程中,组成了一个动物-细菌共同体,进而构建成为一个协调而相对独立的微生态环境。在这一特殊环境中,放线菌所产生的次级代谢产物,既可被动物机体所耐受,又具有抑制特定病原微生物的活性。这类次级代谢产物种类繁多、结构特异,具有抗细菌、真菌、病毒、肿瘤及免疫调节等多种生物活性,具备开发成为新型药物的巨大潜力。本文旨在总结近十年来关于动物来源放线菌的次级代谢产物及其生物学活性的研究进展,为进一步开发新型药物提供参考和研究策略。

Advances in secondary metabolites produced by actinobacteria derived from animal-microbe mutualism and their biological activities

Abstract: Actinobacteria remain to be one of the major sources for new antibiotics, which historically play an essential role in human's fight against infectious diseases. Due to the emergence of resistant pathogenic microorganisms such as bacteria, fungi and viruses, it is imperative to develop new and effective drugs against these pathogens. The symbiotic actinobacteria residing inside the animals are becoming more and more important as a new source for drug discovery, as well as a "hotspot" in the field of microbial medicine. During the long period of evolution, a specific host-microbe mutualism is formed between the symbiotic bacteria and their hosts of animals. In this unique ecosystem, the secondary metabolites produced by bacteria are well tolerated by the hosts, meanwhile, are able to selectively suppress pathogenic microorganisms, thus providing a specific protection to their hosts. These secondary metabolites encompass a large variety of structural diversities of natural products, and so far, the reported biological activities are including antibacterial, antifungal, antiviral, antitumor, and immunomodulatory effects, which give them a great potential in the field of drug discovery. Herein, we review the secondary metabolites of animal symbiotic actinobacteria and their biological activities within the recent decade, by which to provide a viewpoint for future research of drug discovery from actinobacteria.

天然化合物是新药或其先导化合物的重要来源之一。例如, 目前上市的抗肿瘤药物中, 直接或间接来源于天然产物的药物占63%, 这一结果在抗感染药物领域表现得尤其明显, 约占70%[1]。作为天然产物的一个重要组成部分, 放线菌产生的次级代谢产物是其中最活跃的领域之一。自1941年发现链霉素以来, 到目前为止, 据估计已发现的10 000多种抗生素中, 有80%是放线菌产生的。此外, 报道过的22 000多个微生物的次级代谢产物中, 有70%来自放线菌、20%来自真菌、10%来自其他微生物[2]

放线菌(actinobacteria)属于革兰阳性菌, 具有高的G + C含量, 大多数有发达的分枝菌丝, 菌丝纤细, 宽度约0.5~1 μm。作为天然产物资源的一部分, 放线菌在自然界分布很广, 次级代谢产物具有种类复杂性、多样性和特殊性的特点, 为新颖的化合物结构骨架提供了可能, 在天然产物研究中一直占有重要的地位[3]。然而, 经过上世纪几十年的快速发展, 目前已经达到一个瓶颈, 寻找新的资源成为迫切需求。近年来, 越来越多的研究表明, 动物来源的放线菌是微生物药物开发潜力最大的一类微生物资源, 显示出了较好的应用前景。本文将从生物活性的角度总结近十年来关于动物来源放线菌的次级代谢产物及其生物学活性的研究进展, 为进一步深入开发这一资源提供参考。

1 动物来源放线菌的特点

据估计, 地球上生活着数百万种动物, 它们的形态和生活习惯各异, 为体内微生物生长提供了多样的环境。动物来源的放线菌由于其遗传和代谢, 以及与动物相互作用的不同, 造就了不同于外界环境微生物的特性。由于宿主可以对微生物进行选择, 这些微生物进而与宿主形成互惠共生的复合体, 因此动物体内这一环境下微生物的多样性可以说是宿主与相应部位微生物之间相互选择和协同进化的结果。例如宿主的肠道等部位为微生物提供了包括氧气、营养物质、天然屏障、适宜的温度和pH等生长和繁殖所需的各种生理条件, 而微生物反过来影响宿主的能量摄入、分布和消耗等代谢功能, 进而影响宿主的健康[4-6]。研究表明, 这类微生物表现出多种多样的生物学功能, 且属于相对未开发的菌群, 因此值得深入研究和探索[7-9]

截至目前, 人们研究了多种动物体内分离得到的微生物产生的次级代谢产物及其生物活性[10], 这些菌株分别来源于人体[11]、大熊猫[12]和犬[13]的肠道, 以及多种海洋动物体内等, 其中许多研究与放线菌相关。例如从这些特殊环境的微生物中, Ding等[14]从大象肠道放线菌中分离得到多个高活性的抗肿瘤和抗真菌化合物, Viaud等[15]发现肠道微生物在肿瘤免疫疗法方面的巨大潜力, Rea等[16]从人体肠道细菌中分离得到专一性抗难辨梭状芽孢杆菌化合物thuricin CD, 以及Zhao等[17]的研究表明, 肥胖症、糖尿病等疾病也与肠道微生物的某些菌群相关。大量研究结果表明, 动物来源放线菌的次级代谢产物种类繁多、结构新颖, 具有抗细菌、真菌、病毒以及肿瘤等多种多样的生物活性。

2 抗细菌和真菌活性

动物来源放线菌的次级代谢产物中, 具有多种吩嗪类、生物碱类、内酯类以及环肽类化合物。研究发现, 这些化合物具有显著的抗细菌、真菌等活性, 发挥着特定的生物学功能。

海绵作为世界上最原始、最简单的多细胞动物, 种类达到海洋动物种类的1/15, 是一个庞大的多孔类动物“家族”。研究表明, 许多从海绵中分离得到的放线菌菌株具有较好的抗菌活性[18-21]。例如, Karuppiah等[19]从中国南海采集的7种海绵中分离得到的197株放线菌中, 通过基因筛选和化学分析筛选相结合的方法, 分离得到两个具有抗蕈状芽孢杆菌、金葡菌、大肠埃希菌和滕黄微球菌活性的化合物1, 6-dihydroxyphenazine (1)和1, 6-dimethoxyphenazine (2)。此外, Dashti等[20]Actinokineospora sp. EG49和Nocardiopsis sp. RV163两株放线菌的共培养代谢物中也分离得到了上述化合物2。他们发现这个化合物不仅可以抑制芽孢杆菌和布氏锥虫, 还可以抑制Actinokineospora sp. EG49的生长, 由此说明菌种共培养可以激发某些放线菌沉默基因的表达。Ng等[21]发现, 盐孢菌属海洋放线菌能产生利福霉素等生物活性物质从而具有抗结核分枝杆菌的活性。为此, 他们从澳大利亚大堡礁的海绵中分离得到Salinispora arenicola M413, 并探索了该菌合成利福霉素的生物合成途径。

珊瑚是属于刺胞动物门珊瑚虫纲的海生无脊椎动物。Zhang等[22]研究了中国南海浅水珊瑚中123株可培养放线菌的物种多样性及其抗菌活性, 随后Li等[23]研究了中国南海造礁石珊瑚中得到的70株可培养放线菌的抗菌活性, 他发现其中6株放线菌的粗提物显示出与环丙沙星相当甚至更好的抗溶藻弧菌活性。此外, Nong等[24]从寄生于中国南海柳珊瑚(Muricella flexuosa)的放线菌(Streptomyces sp. SCSGAA0009) 中分离到2个生物碱类化合物, N-[2-(1H-indol-3-yl)ethyl]propionamide (3)和phenazine-1-carboxylic acid (4)。其中化合物4对大肠埃希菌和海洋细菌假单胞菌(P. piscida UST010723-006) 具有显著抑制作用。同时, 该化合物还具有抗草苔虫幼虫的附着作用(EC50 < 25 μg·mL-1)。因此可以推断, 化合物4具有抵抗海洋细菌对珊瑚的入侵和致病作用, 以及潜在的天然抗污作用(美国海军舰队规定的天然抗污损剂EC50 = 25 μg·mL-1)。

蚂蚁属于膜翅目蚁科昆虫, 遍布全球, 是一种社会性昆虫, 在热带地区最为常见。日本科学家Matsui等[25]从白蚁的消化道中得到一株具有抗微球菌、芽孢杆菌和假丝酵母菌活性的链霉菌CA1, 并进一步从该菌的代谢产物中纯化得到actinomycin X2 (5)和D (6)两个化合物。化合物5是内酯类抗生素, 而6则具有抗真菌和抗癌活性。从南美的Trachymyrmex属蚂蚁中, Mendes等[26]分离得到24株放线菌, 从中筛选具有抗白色念珠菌活性的菌株。在此基础上, 他们从Streptomyces sp. TD025中分离得到两个新化合物urauchimycins A (7)和B (8)。这些化合物表现出较好的抗白色念珠菌活性(表 1)。Oh等[27]从寄生于蚂蚁(Apterostigma dentigerum)表皮的放线菌(Pseudonocardia sp.)中分离得到一个环状的缩肽类化合物dentigerumycin (9)。令人称奇的是, 该化合物对蚂蚁身上共存的不同属真菌具有选择性的抑制作用。此外, Dangelo等[28]也发现切叶蚁表皮中的放线菌可以抑制切叶蚁身上寄生的丝状真菌, 从而达到保护切叶蚁的作用。

白蚁可以利用自身的粪便作为巢穴的骨架。Visser等[29]推测从荷兰白蚁巢穴中分离得到的放线菌可能能够抑制白蚁的共生真菌PseudoxylariaTermitomyces从而保护白蚁。为此, 他们从白蚁巢穴不同的位置分离出360株放线菌, 然后通过抗菌实验证明了这一猜想。与此相似, Chouvenc等[30]从美国白蚁Coptotermes formosanus巢穴中找到一株放线菌, 它可以在白蚁觅食的时候抑制黑僵菌Metarhizium anisopliae, 进而起到保护白蚁的作用。他们进一步研究了该放线菌株对不同属真菌的抑制活性以及抗黑僵菌的作用机制。此外, Lu等[31]从白蚁巢中分离得到一株放线菌Streptomyces violaceoruber BYC 01, 发现其发酵液的乙酸乙酯提取物能够抑制苹果树腐烂病菌、水稻纹枯病菌、杨树溃疡病菌、白色念珠菌、金葡菌、大肠埃希菌、枯草芽孢杆菌和水稻白叶枯病菌, 并从中分离得到单体化合物fogacin (10)。活性测试结果表明, 该化合物能够抑制白色念珠菌的生长。

从胡蜂体表及体内分离出放线菌株并测试其抗菌活性是另一报道较多的研究[32, 33]。Oh等[34]从寄生于美国威斯康星州胡峰(Sceliphroncae mentarium)体表的放线菌(Streptomyces sp.)中分离得到一个具有9个双键、26个碳原子的大环酰胺类化合物sceliphrolactam (11), 该不饱和化合物性质不稳定, 易被光照、高温和路易斯酸分解。他们发现在避光的条件下, 化合物11对耐两性霉素的白色念珠菌具有显著的抑制活性(MIC = 4 μg·mL-1)。

此外, 还有文献报道了从蚯蚓, 淡水鱼肠道, 北美洲鹑的气管、食管、盲肠和泄殖腔, 以及梅花鹿、大象等中分离出放线菌菌株, 并测试了其抗菌活性[13, 35-38]。其中从我国一级保护动物梅花鹿的皮肤中分离得到的放线菌菌株JN411010, 经培养后纯化得到14个化合物(12~25), 部分化合物对芽孢杆菌、白喉杆菌和大肠埃希菌表现出较好的抑制活性[38]

3 抗肿瘤活性

天然化合物及其衍生物是抗肿瘤化合物的重要来源之一。细胞毒性实验结果表明, 动物来源放线菌的次级代谢产物也具有比较明显的抗肿瘤活性。Motohashi等[39]从海绵中分离得到的放线菌Streptomyces sp. Sp080513GE-26中得到3个具有抗人宫颈癌HeLa细胞和抗人急性骨髓性白血病HL-60细胞活性的化合物, 分别命名为tetracenoquinocin (26)、5-iminoaranciamycin (27)和aranciamycin (28)。

Wyche等[40]从海绵Chondrilla caribensis f. caribensis中分离得到一株疣孢菌, 然后从其代谢产物中分离出5个新的thiocoraline类似物, 其中22'-deoxythiocoraline (29)、12'-sulfoxythiocoraline (30)和thiochondrilline C (31)都具有抗A549肿瘤细胞的活性, 其EC50值分别为0.13、1.26和2.86 μmol·L-1。从海绵中得到的稀有放线菌Amycolatopsis sp.中, Kwon等[41]纯化得到具有较强抗胃癌细胞SNU638和结肠癌细胞HCT116活性的化合物amycolactam (32), 其IC50值分别为0.8和0.2 μmol·L-1

化合物lobophorin C (33)和D (34)是从中国东海海绵里面分离得到的放线菌Streptomyces carnosus AZS17中分离出的两个次级代谢产物, 其中化合物33能够抑制人肝癌细胞7402的生长, 而34对人乳腺癌细胞MDA-MB 435的抗肿瘤活性较高[42]

从海绵里面分离得到的放线菌中, Khan等[43]筛选得到具有抗人宫颈癌HeLa细胞和恶性间皮瘤ACC-MESO-1细胞活性的阿雷西霉素类似物JBIR-43。Vicente等[44]则从波多黎各海绵Scopalina ruetzleri中得到的一株放线菌Streptomyces sp. M7-15中分离得到9个化合物, 其中化合物frigocyclinone (35)、monacyclinone C (36)、monacyclinone E (37)和monacyclinone F (38)对rhabdomycosarcoma肿瘤细胞(SJCRH30) 的抑制活性的IC50值分别为5.2、160、270和0.73 μmol·L-1, 且38还具有抗革兰阳性菌的活性。

Schneemann等[45]通过基因筛选的方法从海绵Halichondria panicea中分离得到的一株放线菌Streptomyces sp. HB202, 并从该菌的发酵液中纯化出一个芳香多酮类化合物mayamycin (39)。活性测试表明, 该化合物对8种不同肿瘤细胞(肝癌细胞HepG2、结肠腺癌细胞HT-29、胃癌细胞GXF251L、非小细胞肺癌细胞LXF529L、乳腺癌细胞MAXF401NL、黑色素瘤细胞MEXF462NL、胰腺癌细胞PAXF1657L和肾癌细胞RXF486L)都具有较好的抑制作用, 并且还可以抑制多种耐药细菌株的生长(表 2)。

海星属于海洋中的一类肉食性棘皮动物, 在海洋碳循环中有着重要作用。Shin等[46]从棘冠海星Acanthaster planci中得到的放线菌次级代谢产物中分离获得4个violapyrones化合物(40~43), 它们对所测试的10株不同肿瘤细胞都有抑制活性, 其GI50值在1.10~26.12μg·mL-1之间(表 3)。

Ye等[47]从海参的泄殖腔中分离得到一株放线菌Pseudonocardia sp. HS7, 并从中纯化得到5种弯孢霉素大环内酯类化合物(44~51), 其中化合物44~4951对神经胶质瘤C6、U87-MF、SHG-44和U251细胞株, 结直肠癌HCT-15、SW620细胞株等均有不同程度的抑制作用, 且化合物47的活性最好(IC50 = 0.59~3.39 μmol·L-1)。此外, 4445还能抑制大肠埃希菌的生长。

从中华稻蝗消化道中分离出的稀有放线菌Amycolatopsis sp. HCa1中, Guo等[48]纯化得到10个化合物(52~61)。经抗肿瘤活性检测表明, 化合物53556061抑制人宫颈癌HeLa细胞的IC50值分别为0.27、0.11、0.56和0.39μmol·L-1

蜣螂俗名屎壳郎, 会收集食草动物的粪便滚成球状, 并在粪球(又称育雏球)中产下一个虫卵, 幼虫在充满微生物的粪球中经数月可孵育长大。蜣螂、幼虫及粪球中的微生物形成了一个共存和谐的整体。Kim等[49]从蜣螂(Copristri partitus)育雏球分离得到的放线菌(Streptomyces sp.)里面得到6个化合物(62~67)。这些化合物具有类似1-甲基-4-苯基-1, 2, 3, 6-四氢吡啶(MPTP)的结构, 因此可代谢为1-methyl-4-phenylpyrimidium (MPP+), 从而具有神经细胞毒性, 引起帕金森病。他们还发现, 化合物62 (IC50 = 5 μmol·L-1)、63 (IC50 = 2.5 μmol·L-1)能显著降低MPP+对多巴胺能神经细胞SH-SY5Y的毒性作用, 推测其吡啶环上的苯基可能是重要的效应基团。此外, 由于化合物64 (IC50 = 27 μmol·L-1)、65 (IC50 = 107 μmol·L-1)母核上存在着吡啶环, 因此具有一定的抗肿瘤活性。

4 其他生物活性

除抗细菌、真菌和肿瘤活性外, 从动物来源的放线菌中还分离得到具有抗病毒、抗氧化、抗寄生虫、抗水解酶, 以及抗疟疾等多种生物活性的化合物。研究表明, 由于这些化合物具有特定的药效基团, 所以表现出特定的生物学活性。

从寄生于热带海洋动物鸡心螺(Conuscir cumcisus)毒液管的放线菌(Gordonia sp. 647W.R.1a.05) 中, Lin等[50]纯化得到12个化合物, 其中4-hydroxysattabacin (68)和soraphinol C 12 (69)能够抑制单纯孢疹病毒(Herpes simplex)。

Grkovic等[51]从海绵中分离得到的Actinokineospora sp. EG49菌株中分离得到两个化合物actinosporins C (70)和actinosporins D (71), 它们具有明显的抗氧化活性, 并且能对由过氧化氢导致的基因损伤提供保护作用。此外, Cheng等[52]从希腊海绵中发现一株放线菌株具有抗锥虫活性, 以及Tabares等[53]从加勒比海绵分离得到的6株放线菌具有抑制蛋白酶和诱导人类外周血中细胞因子释放的活性。

三开蜣螂是一种有名的杂食性害虫。Kim等[54]从三开蜣螂Copris tripartitus来源的链霉菌中分离得到4个对MPP+导致的神经母细胞瘤SH-SY5Y细胞的神经毒性有保护作用的化合物coprismycins A (72)和B (73), 以及dipyridines (74, 75)。Park等[55]从蜣螂(Copristri partitus)育雏球的放线菌中分离得到1个三环内酰胺结构化合物tripartilactam (76), 该化合物具有独特的4-, 8-, 18-三环结构, 在臭氧和酸存在的情况下, 易于分解为3-氨基-2-甲基丙酸的结构。研究发现该化合物具有抑制Na+/K+-ATP酶的作用(IC50 =16.6 μg·mL-1)。

另据报道, Oh等[56]从美国南方松甲虫(Dendroctonus frontalis)的共生放线菌次级代谢产物中分离得到一个多烯过氧化物(77)。该化合物含有7个不饱和双键、1个双氧戊环和1个羧基。77对担子菌类具有较强的抑制能力, 而对子囊菌类则不敏感(表 4)。另外, 该化合物(EC50 = 17 ng·mL-1)还具有类似于青蒿素、氯喹(EC50 = 10 ng·mL-1)的抗疟作用。由此表明, 许多放线菌能够以次级代谢产物为媒介, 帮助宿主对抗外来真菌的感染, 进而造就一个宿主-寄生真菌-细菌三者和谐共存的生态环境[57]

5 构效关系研究

动物来源放线菌的次级代谢产物种类繁多、结构各异。从化合物母核来看, 吩嗪类化合物(124)多具有较好的抗细菌活性, 而大环内酯类化合物(5~7)、多烯类(11)、环状缩肽类(921)等则表现出较高的抗真菌的活性。到目前为止, 分离得到的抗肿瘤化合物的数目最多的, 其结构骨架主要为蒽醌类(如26~2835~3952~61等)、大环内酯类(44~51), 以及部分带有7到8个碳原子饱和脂肪烷烃侧链的吡喃酮类化合物(40~43)。因此, 根据这些次级代谢产物的药效基团, 可在一定程度上推测其所具有的生物活性。例如deoxythiocoraline (29)、sulfoxythiocoraline (30)和thiochondrilline C (31)都具有3-羟基喹哪啶结构, 是噻可拉林类似物[40], 因此这些化合物都具有抗肿瘤细胞的作用。Coprismycin A (62)具有类似1-甲基-4-苯基-1, 2, 3, 6-四氢吡啶(MPTP)的结构, 可减轻内源性MPP+诱导的神经毒性, 具有治疗神经退行性疾病如帕金森氏病的潜力[49]; 再例如mycangimycin (77)具有与青蒿素相类似的1, 2, 4-三氧杂环己烷结构, 因而表现出抗疟原虫的活性[56]

进一步从取代基来看, 相比urauchimycins A (7)与urauchimycins B (8)[26], 二者仅侧链甲基取代位置不同, 但在抗真菌活性上差别较大, 由此表明空间关系对其抗真菌活性有着重要的影响(表 1)。噻可拉林(thiocoraline)能显著抑制人肺腺癌细胞A549的生长(EC50 = 0.009 5 μmol·L-1), 但其22'位脱羟基衍生物deoxythiocoraline (29)活性显著降低(EC50 = 0.13 μmol·L-1); 而当12'位的硫甲基被氧化为具有亚砜结构的sulfoxythiocoraline (30)时, 其抗肿瘤活性进一步降低(EC50 = 1.26 μmol·L-1)[40]。由此推测, 空间效应, 尤其是电子效应能显著改变噻可拉林的抗肿瘤活性。此外, lobophorin C (33) (R = NO2, IC50 = 7.5 μmol·L-1)和D (34) (R = NH2, IC50 = 61.8 μmol·L-1), 由于取代基、立体构型等的不同, 它们对人乳腺癌细胞MDA-MB-435的抑制作用也截然不同[41], 表明这些化合物生物活性与其化学结构之间存在着明显的依赖关系。

6 研究方法和手段的概况

近年来, 从动物来源放线菌的次级代谢产物中寻找具有生物活性先导化合物的研究中, 除了传统的途径外, 还采用了一些新的方法和手段。例如, 通过对次级代谢产物生物合成途径中标志性基因片段进行筛查[19, 58], 从而寻找能够产生特定结构化合物的菌株, 并结合液相色谱—质谱联用的方法, 获得目标化合物, 以及通过与目标致病菌共同孵育培养的方法来激活沉默基因, 进而获得具有特定生物活性的次级代谢产物[20]。此外, 从动物-细菌共同体这种特定环境中去寻找特定的菌株[27, 28, 34, 56], 也得到一些具有较好生物活性和新颖结构的化合物。

7 总结与展望

动物来源的放线菌资源丰富, 数量繁多, 其次级代谢产物大多为氮杂环、芳香类和环肽类化合物, 结构骨架变化较大。从宿主来看, 由于海绵[59, 60]、珊瑚[61, 62]、蚯蚓[63, 64]等具有独特的生活习性和药用价值[65], 长期以来都是研究的热点。在这些研究中, 蚂蚁[66]、蚯蚓等昆虫由于携带不同菌群的机会较高, 相关的文献报道更多一些。近年来, 由于大象[14]、企鹅[67]等高等动物的肠道细菌易于从它们的粪便中分离培养, 且种类丰富, 其次级代谢产物具有毒性低、抗菌谱窄、活性强等特点, 已成为新的热点。

共生细菌寄生于动物的体表或者体内, 在与动物和谐共存的过程中, 构建了一个完整的微生态环境, 共同维持生态的平衡。动物为细菌提供了生存的环境和养分, 细菌则分泌众多次级代谢产物, 帮助宿主抵抗病原菌的侵袭和繁殖。动物-细菌共生体因而成为自然界中一个特殊而稳定的生态微环境。在这一环境下生存的放线菌, 其产生的次级代谢产物不仅在抗菌方面表现突出, 在抗肿瘤、病毒以及寄生虫等方面也表现出巨大潜力。随着耐药性病原微生物的不断出现, 有关动物来源放线菌次级代谢产物及其生物活性的研究将变得越来越重要。这一领域虽然起步较晚, 但发展迅速, 显示了极好的开发和应用前景。
